摘要
目的:探讨维生素E对哮喘大鼠气道结构重塑的影响。方法:24只清洁级SD雄性大鼠随机分为对照组、哮喘组和维生素E组,每组8只。哮喘组和维生素E组用卵白蛋白致敏并激发建立哮喘模型,对照组以等量生理盐水处理;第15天起,维生素E组予维生素E 100 mg/(kg·d)灌胃,其余组用等量生理盐水灌胃;第69天激发后处死大鼠,采集血清及肺组织标本。大鼠肺组织行HE染色观察其病理改变,气管壁及平滑肌厚度经计算机图像分析系统检测,ELISA法测定血清缺氧诱导因子-1α(hypoxia inducible factor-1α,HIF-1α)、基质金属蛋白酶-9(matrix metalloproteinase-9,MMP-9)、基质金属蛋白酶组织抑制因子-1(tissue inhibitor of metalloproteinase-1,TIMP-1),免疫组化法测定肺组织MMP-9、TIMP-1表达。结果:哮喘组气管壁及平滑肌厚度较对照组显著增加;维生素E组上述指标较哮喘组有所减轻;与对照组比较,哮喘组血清HIF-1α、MMP-9、TIMP-1及肺组织MMP-9、TIMP-1明显增加,而维生素E组上述指标较哮喘组显著减少(P <0.05)。结论:维生素E可能经抑制HIF-1α、MMP-9、TIMP-1来改善哮喘气道结构重塑。
Objective:To investigate the effects of vitamin E on airway remodeling in asthma rats. Methods:Twenty-four clean grade SD male rats were randomly divided into the control group,the asthma group and the vitamin E group,with 8 in each. The asthma group and the vitamin E group were sensitized and challenged with ovalbumin to establish asthmatic models. The control group was treated with the same amount of saline. From the 15 th day,the vitamin E group was fed with vitamin E,100 mg/(kg·d). The other two groups were fed with equal amount of saline. On day 69,all rats were sacrificed in 24 hours after challenged,then the serum and lung tissue specimen were preserved. The pathological changes of lung tissues were detected by HE staining. The thickness of airway wall and smooth muscle was detected by computer image analysis system. The expression of hypoxia inducible factor-1α(HIF-1α),matrix metalloproteinase-9(MMP-9)and tissue inhibitor of metalloproteinase-1(TIMP-1)in the serum was detected by ELISA. MMP-9 and TIMP-1 in lung tissues were observed by immunohistochemistry. Results:Compared with the control group,the thickness of airway wall and smooth muscle in the asthma group significantly increased,while the vitamin E group somehow improved in the above changes when compared with the asthma group. Compared with the control group,the expression of HIF-1α,MMP-9,TIMP-1 in serum and MMP-9,TIMP-1 in lung tissues in the asthma group significantly increased. while HIF-1α,MMP-9,TIMP-1 in serum and MMP-9,TIMP-1 in lung tissues in the vitamin E group obviously decreased when compared with the asthma group(P〈0.05). Conclusion:Vitamin E may improve airway remodeling in asthma by inhibiting HIF-1α,MMP-9 and TIMP-1.
