miR-214-3p对肺癌细胞增殖的影响

Effect of miR-214-3p on proliferation of lung cancer cells

目的通过过表达及抑制miR-214-3p研究其对肺癌A549、H1299细胞增殖的影响。方法合成miR-214-3p mimics及其对照,合成miR-214-3p抑制剂序列及其对照序列,分别转染肺癌细胞后,运用实时荧光定量PCR(real-time PCR,RT-PCR)法检测miR-214-3p的表达情况。CCK-8法检测miR-214-3p mimics及miR-214-3p抑制剂对肺癌细胞增殖能力的影响。结果RT-PCR结果表明miR-214-3p mimics转染A549、H1299细胞后miR-214-3p表达水平显著升高(均P <0. 05),miR-214-3p抑制剂转染A549细胞后miR-214-3p的表达降低(P <0. 05),miR-214-3p抑制剂转染H1299细胞后miR-214-3p的表达差异无统计学意义。miR-214-3p mimics转染A549、H1299细胞后,肺癌细胞的增殖能力下降(P <0. 01); miR-214-3p抑制剂转染H1299细胞后,肺癌H1299细胞的增殖增加(P <0. 01)。结论 miR-214-3p可抑制肺癌A549、H1299细胞增殖。

Objective To investigate the effect of nfiR-214-3p on proliferation of lung cancer cells A549,H1299 after over-expression or inhibiting the expression of nfiR-214-3p. Methods The nfiR-214-3p nfinfies and its control, nfiR-214-3p inhibitor sequences and its control sequences were synthesized. The expression level of nfiR-214-3p was deternfined by real-time quantitative PCR. The effects of nfiR-214-3p and nfiR-214-3p inhibitors on the proliferation of A549 and H1299 cells were detected by CCK-8 assay-. Results Re- al-time quantitative PCR result showed that the expression of nfiR-214-3p significantly increased in A549, H1299 cells after transfeeted with nfiR-214-3p nfinfics（P 〈 0. 05） , and the expression of nfiR-214-3p was suppressed in A549 cells after transfected with nfiR-214- 3p inhibitor（P 〈 0. 05 ） , while the expression of nfiR-214-3p showed no significant difference in H1299 cells after transfected with nfiR- 214-3p inhibitor. The proliferation of lung cancer cells decreased in A549,H1299 cells after transfected with miR-214-3p mimics（P 〈 0. 01 ） ,while the proliferation of lung cancer cells was promoted in H1299 cells after transfected with miR-214-3p inhibitor（P 〈 0. 01 ）. Conclusion The miR-214-3p could inhibit the proliferation of lung cancer cells.
