CEACAM-1和MMP-2在结直肠癌中的表达及与淋巴结转移的关系 被引量:8

Expression of CEACAM-1 and MMP-2 in colorectal adenocarcinoma and its correlation to lymph node metastasis

摘要 目的探讨癌胚抗原相关细胞黏附分子1(CEACAM-1)、基质金属蛋白酶2(MMP-2)在结直肠癌中的表达及与淋巴结转移的关系。方法收集河北省吴桥人民医院50例结直肠癌组织和50例正常结直肠组织,用免疫组化法检测CEACAM-1和MMP-2的表达情况,并记录LMVD值,对CEACAM1、MMP-2表达情况与微淋巴管密度(LMVD)关系进行相关性分析。结果CEACAM-1和MMP-2在结直肠癌组织中阳性表达为68. 00%(34/50)和64. 00%(32/50),显著高于正常组织的28. 00%(14/50)和24. 00%(12/50);结直肠癌组织中LMVD值为15. 13±3. 08,明显高于正常组织(5. 15±1. 25); CEACAM-1、MMP-2和LMVD三者均与肿瘤浆膜浸润、TNM分期、脉管浸润、淋巴结转移及肝转移密切相关(均P <0. 01);结直肠癌组织中CEACAM-1和MMP-2阳性组表达组LMVD值为18. 14±3. 71和17. 19±3. 85,显著高于阴性组(10. 20±3. 52和10. 09±3. 08,均P <0. 05);结直肠癌组织中CEACAM-1与MMP-2表达呈明显正相关性(r=0. 489,P <0. 001)。结论 CEACAM-1、MMP-2高表达能诱导肿瘤微淋巴管的生成,与结直肠癌的浸润及淋巴转移有关,通过调控CEACAM-1、MMP-2的表达可能抑制肿瘤的浸润和淋巴结转移。 Objective To explore the the expression of CEACAM-1 and MMP-2 in colorectal cancer and their relationship with lymph node metastasis. Methods Fifty eases of eoloreetal cancer tissues and 50 eases of normal eoloreetal tissues were collected from Wu- qiao People' s Hospital in Hebei Province. The expression of CEACAM-1 and MMP-2 was detected by imnmnohistoehenlistry technique and the lymphatic microvessel density(LMVD) value was recorded. The correlation between the expression and LMVD was analyzed. Results The positive expression rates d CEACAM-1 and MMP~ were 68.00%(34/50) and 64. 00%(32/50) in colorectal cancer tissues, respectively, which were significantly higher than those of normal tissues[28.00%(14/50) and 24.00%(12/50) ]. The LM- VD value was significantly higher in eoloreetal cancer tissues than in normal tissue( 15. 13 ± 3.08 vs 5. 15 ±1.25). The expressions of CEACAM-1, MMP-2 and LMVD were all related to tumor serous menlbrane infiltration, TNM staging, vascular infiltration, lymph node metastasis and liver metastasis( all P 〈 0. 01 ). The LMVD value in eoloreetal cancer tissues was significantly higher in CEACAM-1 positive group than in negative group(18. 14 ±3.71 vs 10. 20 ±3.52,P〈0. 05) , and it was also higher in MMP-2 positive group than in negative group( 17. 19 ± 3.85 vs 10. 09 ± 3.08 ,P 〈 0. 05). The expression of CEACAM-1 was positively correlated with MMP-2 expression in colorectal cancer tissues( r = 0.489,P 〈 0. 001 ).Conclusion The high expression of CEACAM-1, MMP-2 could induce the formation of tumor lymphatic vessels, and be associated with invasion and lymph node metastasis of eoloreetal cancer. Regulating the expression of CEACAM-1, MMP-2 could inhibit tumor invasion and lymph node metastasis.
