桃红四物汤对UV辐照致HDMEC损伤后VCAM-1和bFGF表达的影响

摘要 目的:探讨桃红四物汤对模拟日光中UV辐照所致人真皮微血管内皮细胞(HDMEC)损伤的干预作用。方法:研究以人真皮微血管内皮细胞为研究对象,采用UVA照射的方法造模,以不同浓度的桃红四物汤为干预因素,分别予高、中、低剂量桃红四物汤含药血清培养,用ELISA法检测培养上清液中VCAM-1和bFGF的含量。结果:与空白对照组比较,模型对照组和桃红四物汤中、低剂量含药血清组中的VCAM-1含量均显著上调且有统计学差异(P<0.05,P<0.01),而桃红四物汤高剂量组无统计学差异(P>0.05);与模型对照组比较,桃红四物汤高剂量、中剂量和低剂量含药血清组中的VCAM-1含量均显著下降,存在统计学差异(P<0.01)。与空白对照组比较,模型对照组、桃红四物汤高剂量、中剂量和低剂量含药血清组中的bFGF含量均显著上调且有统计学差异(P<0.01);与模型对照组比较,桃红四物汤高剂量、中剂量和低剂量含药血清组中的bFGF含量均显著下降且有统计学差异(P<0.01)。结论:UVA辐射致HDMEC损伤发生炎症反应,促进VCAM-1、bFGF的表达上调,桃红四物汤干预后二者表达下调,表明桃红四物汤能修复受损的血管内皮细胞,进而降低紫外线辐射造成的皮肤损伤。 Objective: The purpose of this study was to investigate the intervention effect of Taohong Siwu decoc- tion on human dermal microvascular endothelial cell(HDMEC) injury induced by UV irradiation in simulated sunlight. Method: To take HDMEC as the object of the study and adopt the UVA radiation modeling method, with different con- centrations of Taohong Siwu decoction as interference factors, respectively for the high, medium and low dose of Tao- hong Siwu decoction drug-containing serum culture. To detect the content of VCAM 1 and bFGF in serum with ELISA method. Results: Compared with the blank control group, the content of VCAM 1 in model control group, medium and low dose of Taohong Siwu decoction group increased obviously with a significant difference(P〈0.05, P〈0.01); whereas the high dose of Taohong Siwu decoction group had no significant difference compared with other groups(P〉0.05).; Compared with the model control group, the content of VCAM 1 in high, medium and low dose of Taohong Siwu decoc- tion group decreased obviously with a statistic difference(P〈0.01); Compared with the blank control group, the content of bFGF in model control group, high, medium and low dose of Taohong Siwu decoction group increased obviously with a statistic difference (P〈0.01); Compared with the model control group, the content of bFGF in high, medium and low dose of Taohong Siwu decoction group decreased obviously with a statistic difference(P〈0.01). Conclusion: UVA radia- tion induced inflammatory response of HDMEC injury and promoted expression increase of VCAM 1 and bFGF. After the intervention of Taohong Siwu decoction, both indicators decreased. The study shows that Taohong Siwu decoction could repair damaged endothelial cells, thus reducing skin damage caused by UV radiation.
