高温对犬血清急性期反应标志物及氧化还原状态的影响 被引量:2

EFFECTS OF HIGH TEMPERATURE ON SERUM ACUTE PHASE REACTION MARKERS AND REDOX STATUS IN DOGS

摘要 目的以犬为实验动物研究急性短期高温环境下血清急性期反应标志物及氧化还原指标的变化,揭示高温环境对犬急性期反应和机体氧化还原状态的影响。方法将实验犬舍温度由(28±1)℃迅速升至(35±1)℃,持续16h后自然降温16 h。试验开始(0h)、1、3、8、16、32 h颈静脉采血2 ml,测定血清中急性期反应标志物和氧化还原指标的活性或含量。结果与试验0h相比,试验3h时,血清抑制羟自由基能力、VC、GSSG、MDA、8-OHdG含量、GSH-Px、CAT活性显著增加(P<0.05),H2O2、羰基含量显著降低(P<0.05);试验8h时铜蓝蛋白活性、T-GSH、羰基含量显著降低(P<0.05),T-AOC、GSH-Px活性、VC、8-OHdG含量显著升高(P<0.05)。试验16h时,血清CRP、HSP70、VC、8-OHdG含量显著升高(P<0.05),铜蓝蛋白、SOD、GSH-Px和CAT活性、-SH、T-GSH和总羰基含量、总抗氧化能力显著降低(P<0.05);试验32h时,铜蓝蛋白、T-SOD、GSH-Px、CAT活性、-SH、H2O2和羰基含量显著降低(P<0.05)、VC、8-OHdG含量显著升高(P<0.05)。结论急性短期高温环境可显著改变血清中VC、GSH-Px、MDA等抗氧化和氧化物质的含量,表明高温环境显著影响了机体体内氧化还原平衡。 Objective This study was performed to determine the changes of serum acute-phase reaction markers and redox indies in dogs exposed to acute short-term high temperature environment, and to reveal the characteristics of acute phase reaction and its effects on the redox status. Methods The temperature of the kennels rose rapidly from (28±1)℃ to (35± 1)℃ for 16h, and then naturally cooled for 16h. At the times 0, 1, 3, 8, 16, 32h, post-experiment blood about 2ml was collected from the jugular vein and determine the activity or content of the acute phase reaction markers and redox indices in serum. Results Compared with Oh, scavenging capacity of hydroxyl radicals, contents of VC, GSSG, MDA, 8-OHdG, activities of GSH-Px and CAT were increased significantly (P〈0.05), and contents of H202 and carbonyl group decreased significantly (P〈0.05); At 8h post-expertiment, contents of ceruloplasmin T-GSH and carbonyl group were decreased significantly (P〈0.05). Meanwhile, T-AOC, activity of GSH-Px and the contents of VC and 8-OHdG were increased significantly (P〈0.05). At 16h post-expenment, the contents of CRP, HSP70, VC and 8-OHdG were significantly increased (P 〈0.05), and the activities of SOD, GSH-Px and CAT, the contents of ceruloplasmin, -SH, T-GSH and carbonyl group, T-AOC decreased significantly (P〈0.05). At 32h post-experiment, the activities of T-SOD, GSH-Px, CAT, the contents of ceruloplasmin,-SH, H202 and carbonyl group were significantly decreased and contents of vitamin C and 8-OHdG were significantly increased (P〈0.05). Conclusion Acute short-term high temperature environment change the contents of antioxidant and oxidative substances significantly such as VC, GSH-Px and MDA in serum, indicateing that the high temperature environment has a significant effect on redox status .
