准噶尔乌头中3种生物碱对H9c2细胞的毒性研究 被引量:2

摘要 目的研究准噶尔乌头中3种生物碱(乌头碱、准噶尔乌碱、12-表-欧乌碱)对H9c2细胞的毒性作用。方法将H9c2细胞分为空白对照组、乌头碱组、准噶尔乌碱组、12-表-欧乌碱组,采用Cell Counting Kit-8(CCK8)法检测准噶尔乌头中3种生物碱对体外对数生长期H9c2细胞的抑制作用,计算细胞增殖抑制率、半数抑制浓度(IC50);乳酸脱氢酶(LDH)漏出率对H9c2细胞的毒性作用;流式细胞术检测准噶尔乌头3种生物碱对H9c2细胞的总凋亡率。结果与空白组比较,乌头碱组、准噶尔乌头碱组、12-表-欧乌头碱组,细胞增殖抑制率指标升高,LDH漏出率指标升高,凋亡率指标升高,差异有统计学意义(P <0.05);对H9c2细胞的毒性作用(乌头碱>准噶尔乌碱>12-表-欧乌碱),IC50值分别为562.06、726.80、766.11μg/mL。结论乌头碱、准噶尔乌碱、12-表-欧乌碱3种生物碱对H9c2细胞具有毒性作用。 Objective To evaluate the toxic effects of three alkaloids (aconitine, Junggar aconitine, 12-epiuridine) extracted in Junggar aconitum on H9c2 cells. Methods H9c2 cells were divided into four groups: blank control group, aconitine group, Junggarine group and 12-epiuridine group. The inhibitory effects of three alkaloids in Junggar aconitum on H9c2 cells in logarithmic phase in vitro were detected by Cell Counting Kit-8 (CCK8) method. The inhibitory rate of cell proliferation, half inhibitory concentration (IC50), the toxic effect of lactate dehydrogenase (LDH) leakage rate on H9c2 cells and the total apoptosis rate of three alkaloids in Junggar aconitum on H9c2 cells were detected by flow cytometry. Results Compared with the blank group, aconitine group, Junggar aconitine group, 12-epiaconitine group, the cell proliferation inhibition rate index significantly increased, the LDH leakage rate index significantly increased, the apoptosis rate index remarkably increased (P 〈0.05). The toxicity to H9c2 cells (aconitine 〉 Junggar aconitine 〉12-epiuridine), the IC50 value was 562.06, 726.80, 766.11 μg/mL, respectively. Conclusion/ Three alkaloids of aconitine, Junggar and 12-epiuridine have toxic effects on H9c2 cells.
