摘要
目的研究不同剂量复方降糖三号方对糖尿病小鼠的治疗作用。方法取昆明种小鼠60只,除空白对照组(蒸馏水,0.1mL/10g体质量)10只外,其余50只采用腹腔注射链脲佐菌素方法(第1天12mg/10g体质量,第3天18mg/10g体质量)建立糖尿病小鼠模型,将造模成功的糖尿病小鼠随机分为模型组、阳性组(参芪颗粒0.1mL/10g体质量)、复方降糖三号方低剂量组(0.2mL/10g体质量)、复方降糖三号方中剂量组(0.4mL/10g体质量)、复方降糖三号方高剂量组(0.8mL/10g体质量)。连续灌胃给药4w后,尾部采血测定空腹血糖(FBG),颈动脉采血测定糖化血红蛋白(HbAlc),将血液离心分离出血清,采用ELISA法测定血清胰岛素(INS),测定小鼠血清胰岛素抵抗指数(HOMA-IR)、血清肿瘤坏死因子α(TNF-α)、核因子-kB(NF-kB)。结果与模型组比较,复方降糖三号方各剂量组小鼠FBG降低,差异有统计学意义(P <0.01);复方降糖三号方高剂量组小鼠INS含量升高,差异有统计学意义(P <0.05);复方降糖三号方各剂量组小鼠胰岛素抵抗指数降低,差异具有统计学意义(P<0.01);高剂量组小鼠HbAlc水平降低,差异有统计学意义(P <0.05);中,高剂量组TNF-α降低,差异有统计学意义(P <0.01);各剂量组NF-kB降低,差异有统计学意义(P <0.01)。结论复方降糖三号方对糖尿病小鼠具有一定的治疗作用。
Objective To observe the therapeutic effect of different doses of compound hypoglycemic 3 on diabetic mice. Methods 60 kunming mice were divided into two groups, with 10 in the blank control group (distilled water,0.1 mL/10 g ) and 50 in experimental group where the 50 were subjected to intraperitoneal injection with cephalosporins method (according to the concentration of 12 mg/10g intraperitoneal injection at the first day, and the concentration of 18mg/10g at the third day) diabetic mice model was successfully established which were randomly divided into model group, the positive group (administered with ginseng and astragalus particles,0.1 mL/10 g), combination hypoglycemic group (0.2 mL/10 g), low dose group compound hypoglycemic dose in the 3rd party group (0.4 mL/10 g), compound hypoglycemic 3rd party high dose group (0.8 mL/10 g). Lavage for four 4 w, after the expiration of the tail abdominal blood test blood sugar, carotid artery blood determination of glycosylated hemoglobin (HbAlc), the centrifugal separation of blood serum, serum insulin (Ins) was evaluated by ELISA method, the serum indexes of insulin resistance (HOMA-IR) and serum tumor necrosis factor alpha (TNF-alpha), nuclear factor kB (NF-kB) were calculated. Results Compared with the model group, the level of compound hypoglycemic number 3 was high, and the blood glucose was significantly decreased in the medium and low dose group (P 〈0.01). The levels of Ins were significantly increased in the high-dose group (P 〈0.05).The insulin resistance index of mice in each dose group significantly decreased (P 〈0.01). The level of HbAlc was significantly decreased in the high-dose group (P 〈0.05). TNF-α in the medium and high dose groups significantly decreased (P 〈0.01). NF-kB significantly decreased in each dose group (P 〈0.01). Conclusion/ Compound hypoglycemic no. 3 was shown to be able to improve the treatment of diabetic mice.
