摘要
目的:优化柳蘑菌丝体高产多糖发酵条件并对其多糖组分进行分析。方法:通过单因素实验,研究碳源、氮源、生长因子对柳蘑菌丝体和多糖含量的影响,并采用正交试验优化其培养基配方。进一步通过单因素实验研究温度、接种量、pH和发酵时间对柳蘑菌丝体的影响,结合正交试验确定最佳发酵条件,并通过气相色谱法分析其多糖的组成成分。结果表明,柳蘑菌丝体发酵最优培养条件为葡萄糖2.5%,蛋白胨1%,VB20.006%,硫酸镁0.1%,温度25℃,接种量15%,p H4,发酵时间10 d,在此条件下,柳蘑菌丝得率为(11.50±0.29) g/L,多糖含量达到(1.330±0.035) g/L。气相色谱分析结果表明,柳蘑菌丝体分别由L-鼠李糖、L-阿拉伯糖和D-半乳糖组成。结论:本研究条件能够提高菌丝体多糖产量,可作为柳蘑菌丝体高产多糖的发酵条件。
Objective: To optimize the fermentation conditions for high-yielding polysaccharide and analyze the polysaccharide composition from mycelia of Pholiota adipose. Methods: The effects of carbon source,nitrogen source and growth factor on the mycelia and polysaccharide content of Pholiota adipose were studied by single-factor experiments,and the medium formulation was optimized by orthogonal test.The effects of temperature,inoculation volume,pH,fermentation time of mycelia were further studied,the fermentation conditions were optimized by orthogonal test,the constituents of the polysaccharides were analyzed by chromatographic.Results showed that,the optimal conditions fermentation of high-yielding Pholiota adiposa mycelia were glucose 2.5%,peptone1%,VB20.006%,Mg SO40.1%,temperature 25 ℃,inoculation amount 15%,pH4,fermentation time 10 d,under these conditions,the yield of mycelia was( 11.50 ± 0.29) g/L,and the yield of polysaccharide was( 1.330 ± 0.035) g/L.In addition,the mycelia of the Pholiota adiposa was composed of L-rhamnose,L-arabinose,and D-galactose.Conclusion: This condition could improve the yield of mycelia polysaccharide and could be used as the fermentation conditions of high-yielding polysaccharide from mycelia of Pholiota adipose.
