miRNA-17-5p在动脉粥样硬化小鼠中表达变化的研究

摘要 目的观察miRNA-17-5p在动脉粥样硬化(As)疾病中的表达变化。方法将10只C57BI/6小鼠作为正常对照组,10只ApoE-/小鼠作为高胆固醇喂养组。正常对照组正常饮食喂养小鼠12周。高胆固醇喂养组采用高胆固醇饮食喂养小鼠12周,然后尾静脉注射miRNA-17_5p抑制剂干预。观察比较HE染色检测各组小鼠动脉血管的病理变化,miRNA-17-5p在外周血淋巴细胞中的表达和在主动脉血管组织中的表达变化。结果高胆固醇喂养组与正常对照组比较,血管内皮有明显的斑块形成,平滑肌细胞排列紊乱,内膜增生严重;当给予miRNA-17-5p拮抗剂干预后,与高胆固醇喂养组干预前相比,病变程度显著减轻。外周血分离淋巴细胞进行实时荧光定量分析显示,高胆固醇喂养组miRNA-17.5p表达量(0.292±0.062)显著高于正常对照组(0.167±0.016),差异有统计学意义(P〈0.01)。当给予miRNA-17-5p拮抗剂干预后,miRNA-17-5p表达量(0.217±0.023)显著低于高胆固醇喂养组(0.292±0.062),差异有统计学意义(P〈O.01)。高胆固醇喂养组miRNA—17-5p表达量(0.787±0.102)显著高于正常对照组(0.307±0.207),差异有统计学意义(P〈0.01)。当给予rnjRNA-17-5p拮抗剂干预后,miRNA-17-5p表达量(0.535±0.209)显著低于高胆固醇喂养组(0.787±0.102),差异有统计学意义(P〈0.01)。结论miRNA-17-5p在动脉粥样硬化疾病中表达显著升高,可能为临床早期诊断、治疗及危险评估提供理论依据和治疗指导,有望成为动脉粥样硬化新的诊断方式和治疗靶点。 Objective To investigate the expression change of miRNA-17-5p in atherosclerotic diseases. Methods A total of 10 C57BL/6 mice were in the normal control group, and 10 ApoE-/- mice were in the high-cholesterol feeding group. Mice of the normal control group were fed on normal diet for 12 weeks. Mice of the atherosclerosis group were fed on a high-cholesterol diet for 12 weeks, followed by cau- dal intravenous injection of miRNA-17-Sp inhibitor intervention. The pathological changes of arterial vessels in mice in each group were observed and compared by HE staining, and the expression changes of miRNA- 17-5p in peripheral blood lymphocytes and in aortic vascular tissues were observed. Results Compared with the normal control group, the atherosclerosis group had obvious plaque formation, smooth muscle cells were disordered ,and the intimal hyperplasia was serious. When the miRNA-17-5p antagonist was administered, the degree of the disease was significantly reduced compared with the high-cholesterol feeding group. Real-time fluorescence quantitative analysis of peripheral blood separated lymphocytes showed that the expression of miRNA-17-Sp in the atherosclerosis group( 0. 292± 0. 062 )was significantly higher than that in the normal control group( 0. 167±0. 016 ), with statistically significant differences( P 〈 0. 01 ). After the intervention of miRNA-17-5p antagonist, the expression level of miRNA-17-5p(0. 217 ±0. 023 )was significantly lower than that of the atherosclerosis group ( 0. 292 ± 0. 062), and the difference was statisticallysignificant(P 〈0. 01 ). The expression of miRNA-17-5p in the atherosclerosis group(0. 787 -+0. 102)was significantly higher that in the normal control group (0. 307 ± 0. 207 ), with statistically significant dift^rences( P 〈 0. 01 ). Atter the intervention of miRNA-17-Sp antagonist, the expression level of miRNA- 17-5p(0. 535 + O. 209)was significantly lower than that of the atherosclerosis group( 0. 787 + 0. 102 ) , with statistically significant differences(P 〈 0. Ol ). Conclusion The expression of miRNA-17-Sp is significantly increased in atherosclerotic illness,and it may become a new diagnose method and therapeutic target of atherosclerosis diseases.
