五种血细胞分离机采集外周血单个核细胞的献血者体验、采集效率及产品纯度初步比较

Preliminary comparison of user experience,acquisition efficiency and product purity during PBMC collection with five different blood cell separators

目的比较MCS＋、COBE Spectra、Amicus、COM. TEC和Spectra Optia 5种血细胞分离机在采集健康人外周血单个核细胞（peripheral blood mononuclear cells,PBMC）过程中,每种机器的采集效率、产品纯度以及用户体验。方法自2014年11月20日—2016年4月25日,献血者被分成5组,每组12例,分别使用5种血细胞分离机进行PBMC的采集。所有献血者均是定期捐献单采血小板的固定献血者。所有献血者采集前不使用动员剂。在采集过程中,护理人员密切观察献血者的状态。60例献血者的采集时间为2—3 h,总循环血量为5—8 L,终产品体积为100—120 mL。采集完成后,所有的终产品进行五分类血常规检测。根据公式,计算不同分离机采集PBMC的效率和采集产品纯度,然后进行比较。（采集效率=产品PBMC数/单采全程处理PBMC数;单采全程处理PBMC数=[（采前血常规PBMC浓度＋采后血常规PBMC浓度）/2]×（处理总循环血量-ACD量）;产品PBMC纯度=产品PBMC数/产品白细胞总数）结果经过方差分析,5种血细胞分离机采集效率比较,F=1. 805,P〉0. 05,差异无统计学意义; 5种血细胞分离机产品纯度比较,F=8. 795,P〈0. 05,差异有统计学意义。另外,使用5种机型的献血者均未出现献血反应。结论 5种血细胞分离机PBMC采集效率差异不大,产品纯度有明显差异。献血者使用不同血细胞分离机均无明显献血反应。

Objective To compare the collection efficiency,product purity and user experience of PBMC collection between 5 different blood cell separators,including MCS ＋,COBE Spectra,Amicus,COM. TEC and Spectra Optia. Methods From November 20,2014 till April 25,2016,60 blood donors took part in the experiment and they were divided into five groups. For each group,a single kind of blood cell separator was used. During the collection,the state of blood donors was closely observed. Products were tested by blood routine test,and then collection efficiency and product purity were calculated.Results For collection efficiency,F = 1. 805,P〈0. 05,the difference was not statistically significant. As for the product purity,F = 8. 795,P〈0. 05,the difference was statistically significant. All blood donors showed no obvious adverse reaction.Conclusion There was no significant difference of collection efficiency between different separators. While,as for the product purity,there was significant difference. Donors using different blood cell separators had no obvious blood donation reaction.
