摘要
目的:探讨刺芒柄花素对胶质瘤细胞增殖和迁移作用。方法:通过MTT实验检测刺芒柄花素对胶质瘤细胞(U87和U251)增殖的影响。用流式细胞仪检测刺芒柄花素对U87细胞周期的作用。通过Western blot实验和Real time PCR实验检测刺芒柄花素对Cyclin D1的作用。Transwell实验检测刺芒柄花素对U87细胞迁移的影响。Western blot实验和Real time PCR实验检测刺芒柄花素对MMP2/9的作用。结果:10μmol/L和20μmol/L刺芒柄花素对细胞作用24 h后对U87细胞增殖抑制率分别是(26. 74±4. 26)%和(34. 74±2. 40)%;对U251细胞增殖的抑制率分别为(10. 14±9. 80)%和(27. 16±9. 21)%。流式实验指出刺芒柄花素对U87细胞的G1-S期具有阻滞作用。Western blot和Real time PCR实验指出刺芒柄花素可以通过抑制Cyclin D1的表达来抑制U87细胞的增殖。接下来实验发现刺芒柄花素通过抑制MMP2/9来抑制U87细胞迁移。结论:刺芒柄花素可以通过抑制Cyclin D1和MMP2/9来抑制胶质瘤细胞的增殖和迁移。
Objective:To investigate the inhibitory effect of Formononetin on the proliferation and migration of glioma cells.Methods:The proliferation of glioma cells (U87 and U251) was detected by MTT assay after treated by different concentrations of Formononetin.Cells cycle was tested by flow cytometry.The effect of Formononetin on Cyclin D1 was examined by Western blot and Real time PCR assay.The effect of Formononetin on migration of U87 cells was examined by Transwell.The effects of Formononetin on MMP2 and MMP9 were examined by Western blot and Real time PCR assay.Results:After 10 μmol/L and 20 μmol/L of Formononetin were treated on U87 cells,the inhibition of proliferation were (26.74±4.26)% and (34.74±2.40)%. After 10 μmol/L and 20 μmol/L of Formononetin were treated on U251 cells,the inhibition of cell proliferation were (10.14±9.80)% and (27.16± 9.21)%.Flow cytometry showed that the block of Formononetin on the G1-S phase of U87 cells.Formononetin could inhibit the proliferation of U87 cells by inhibiting the expression of Cyclin D1,inhibit the migration of U87 cells by inhibiting the expression of MMP2/9.Conclusion:Formononetin can inhibit the proliferation and migration of glioma cells by inhibiting Cyclin D1 and MMP2/9.
