去铁胺对HL-60细胞Nix蛋白表达的影响

摘要 目的探讨去铁胺(DFO)对HL-60细胞Mix蛋白表达的影响。方法应用不同浓度的DFO刺激HL-60细胞系12 h后采用western blot方法检测处理后细胞中Nix蛋白的表达水平。结果与对照组(0μmol/L)相比,20μmol/L DFO组Nix蛋白表达水平无明显改变(P>0.05),100μmol/L DFO组Nix蛋白表达量明显增加(P<0.01)。结论 DFO可能通过诱导Nix蛋白的表达来实现HL-60细胞的凋亡。 Objective Our previous studies have confirmed that DFO can induce apoptosis in HL-60 cells, but it is not clear whether it is related to the induction of Nix protein. This study is intended to explore the role of Nix protein in the process of mitochondrial autophagy and apoptosis induced by HL-60 cells. Methods Different concentrations of DFO were used to stimulate HL-60 cell line for 12 h and then western blot method was used to detect the expression of Nix protein in the teated cells. Results Compared with the control group (0 μmol/L) , there was no significant change in the expression of Nix protein in the 20 μmol/L DFO group (P 〉0. 05), while the expression of Nix protein in the 100 μmol/L DFO group was significantly higher ( P 〈 0.01 ). Conclusions DFO may promote apoptosis of HL-60 cells by inducing Nix protein.
