肝郁证元素在建立多囊卵巢大鼠模型中的干预作用

Intervention Effect of Liver Depression Syndrome Element on Establishing Polycystic Ovary Rat Model

目的:探讨比较单纯使用"肝郁"、雄激素和"肝郁"联合雄激素3种造模方法对青春期多囊卵巢综合征(PCOS)大鼠模型的影响。方法:采用单纯给大鼠"肝郁"刺激、单纯皮下注射脱氢表雄酮(DHEA)和使用"肝郁"刺激联合皮下注射脱氢表雄酮(DHEA)3种方法建立模型,观察大鼠动情周期和病理组织形态学以判定动物造模状况;采用自动分光光度法测定PCOS大鼠甘油三脂(TG)、总胆固醇(TC)、高密度脂蛋白(HDL)、低密度脂蛋白(LDL),采用化学发光免疫吸附法测定胰岛素(INS),采用酶联免疫吸附法测定瘦素(LP)、脂联素(ADP)的相应变化。结果:肝郁加雄激素组与正常对照组比较,TG、TC、LDL、LP值显著升高,ADP值显著降低;肝郁加雄激素组与雄激素组比较,TG、LDL明显升高,ADP值明显降低。结论:提示"肝郁"使雄激素致青春期多囊卵巢综合征大鼠脂质代谢紊乱程度加重。

Objective： Comparison of the effects of three kinds of modeling method on the use of＂liver depression＂,androgen,androgen combined with ＂liver depression＂ on lipid metabolism in rats with adolescent polycystic ovary syndrome（PCOS）.Methods：Three kinds of rat models were established by ＂Liver Depression＂ stimulation, subcutaneously injecting dehydroepiandrosterone（DHEA）, using ＂Liver Depression＂ stimulation combined with subcutaneous injection of dehydroepiandrosterone（DHEA）.The animal model was determined by observing the estrous cycle and pathological morphology of the rats; the levels of triglyceride（TG）, total cholesterol（TC）, high density lipoprotein（HDL） and low density lipoprotein（LDL） in serum of PCOS rats were determined by automatic spectrophotometry; the levels of insulin（INS） was determined by chemiluminescent; the levels of immunosorbent Leptin（LP） and adiponectin（ADP） were determined by enzyme-linked immunosorbent assay（ELISA）.Results：Liver depression with androgen group compared with normal control group, TG, TC, LDL, LP were significantly increased, ADP was decreased significantly;Liver depression with androgen group compared with androgen group, TG, LDL were increased significantly, ADP was significantly decreased.Conclusions：It is suggested that ＂liver depression＂ can increase the androgen-induced puberty in polycystic ovary syndrome in rats lipid metabolism disorder.
