心康冲剂对慢性心衰大鼠心肌微血管内皮细胞保护作用研究 被引量:8

摘要 目的:研究心康冲剂对心衰大鼠心肌微血管内皮功能保护作用。方法:各组大鼠实验完毕后行心脏超声、血流动力学检测,心脏切片行HE及免疫荧光染色,采用ELISA法检测血清大鼠内皮素-1(ET-1)、血管性血友病因子(vWF)、血小板内皮细胞黏附分子1(CD31)水平。结果:与正常组比较,模型组大鼠心功能明显下降,HE染色示血管形态轮廓模糊,免疫荧光染色示大量荧光点分布,ET-1、vWF、CD31水平升高。与模型组比较,低、中剂组、缬沙坦组心功能增强,HE染色示血管形态形态轮廓较清晰,免疫荧光染色示少量荧光点分布,ET-1、vWF、CD31水平下降。与缬沙坦组比较,高剂量组ET-1、vWF水平升高。结论:心康冲剂低、中剂量能保护心肌微血管内皮功能抗心衰。 Objective:To study the protective effect of Xin Kang Granule(XKG) on myocardial microvascular endothelial function.Method After the experiment,the rats were examined by echocardiography and hemodynamics,the heart slice was stained by HE and immunofluorescence staining.The serum levels of ET-1, vWF and CD31 were detected by ELISA.Results Compared with the normal group,the cardiac function of rats in model group decreased obviously.HE staining showed that the vascular morphology,fuzzy contour fusion, immunofluorescence staining showed a fluorescence distribution,serum levels of ET-1,vWF and CD31 elevated.Compared with the model group,the cardiac function of rats in increased significantly,HE staining showed that the vascular morphology with a clear contour,immunofluorescence staining showed a fluorescence distribution,serum levels of ET-1,vWF and CD31 decreased.Compared with valsartan group,serum levels of ET-1 and vWF in the high dose group increased.Conclusion: Low and middle dose group can protect myocardial microvascular endothelial function and anti heart failure.
