刺老苞根皮醇提物对原代成骨细胞增殖、分化和矿化的影响 被引量:3

摘要 目的:研究4种不同浓度刺老苞根皮醇提物对原代成骨细胞的增殖、分化及矿化的影响。方法:通过消化10只雄性新生SD乳鼠(SPF级)的颅盖骨,获得原代前成骨细胞体外培养、传代,10 mmol/Lβ-甘油磷酸钠和50μg/mL抗坏血酸诱导,碱性磷酸酶(ALP)染色和矿化结节染色鉴定原代成骨细胞,分别用低、中、高浓度(1、10、100μg/mL)刺老苞根皮醇提物(4种)对成骨细胞进行干预,培养4 d和14 d后分别进行ALP活性测定和茜素红染色面积测定。结果:与对照组比较4种刺老苞根皮醇提物均能提高成骨细胞生存率,增强ALP活性,增加矿化结节面积。结论:4种刺老苞根皮醇提物可通过促进成骨细胞增殖和分化,加快骨基质矿化过程。 Objectives: The influence of different concentration groups of 4 kinds ethanol extracts from Aralia echinocauIis Hand.-Mazz. on the proliferation, differentiation and mineralization of primary osteoblasts were observed. Methods: The calvarias of 10 male SD neonatal rats were digested to obtain primary preosteoblasts in vitro subculture. The preosteoblasts were induced to osteoblasts by 10 mmol/L β-glycerin phosphate and 50 μg/mL L-ascorbic acid. Alkaline phosphatase(ALP) and mineralized nodule staining were used to identify primary osteoblasts. The osteoblasts were respectively cultured with 4 kinds of alcohol extracts(1, 10, 100 g/mL) from Aralia echinocauIis Hand.-Mazz. After 4 and 14 days, the activity of ALP and alizarin red staining were measured respectively. Results: compared with the control group, the 4 kinds of alcohol extracts from Aralia echinocauIis Hand.-Mazz. could increase the survival rate of osteoblast, the activity of ALP and the mineralized area. Conclusion: 4 kinds of alcohol extracts from Aralia echinocauIis Hand.-Mazz. can promote the proliferation and differentiation of osteoblasts. Then, increasing the mineralization of bone matrix.
