摘要
以甘薯渣为原料,对米根霉PW352固态发酵产L(+)-乳酸的发酵条件进行了研究。采用单因素试验、均匀设计、正交设计等方法确定最优固态发酵培养基组成为:干薯渣10g/250ml三角瓶、(NH4)2SO41. 5%、MgSO40. 2%、Zn SO40. 05%、MnCl20. 05%、葡萄糖0. 5%、吐温80 0. 05%,在接种量为1. 0×107个孢子/10g干培养基、湿度70%、中和剂碳酸钙30%、36℃条件下,发酵72h产L(+)-乳酸达38. 36(±0. 51)%(w/w),对淀粉糖的转化率为62. 5%。
Production of L( + ) -lactic acid by Rhizopus oryzae PW352 in the solid-state fermentation of sweet potato pulp was investigated by means of orthogondl design, uniform design. A growth condition with 10g sweet potato pulp/250ml flask, ( NH4) 2SO4 1.5% ,MgSO4 0.2% ,ZnSO40.05% ,MnCl2 0. 05% ,glucose 0.5% ,Tween 80 0.05% ,CaCO3 30% , at moisture content of 65% - 70% ,volume of seeds 1.0 × 10^7and 36℃ was favourable for lactic acid fermentation, resulting in lactic acid production of 38.36( ±0.51 ) % (w/w) produced in 72 h fermentation.
