摘要
目的:研究中药肾毒宁方对5/6肾切除慢性肾衰大鼠的疗效作用,并探讨其肾保护作用。方法:将120只SD大鼠随机分为假手术组20只和手术组100只,手术组行5/6肾切除术。术后2周,手术组随机分为病理模型组、中药低、中、高剂量组及西药科素亚对照组,各组大鼠18只。分别给予生理盐水、氯沙坦钾片及相应剂量中药灌胃。治疗2个月后处死大鼠,测定大鼠血肌酐、血尿素氮、血清IL-1β水平以及肾组织超氧化物歧化酶(SOD)、丙二醛(MDA)及肾组织中IV-C表达。结果:与假手术组比较,模型组血肌酐及血尿素氮均明显上升(P <0.05),血清IL-1β水平亦明显升高(P <0.05),SOD明显下调及MDA明显上升(P <0.01),肾组织中IV-C表达亦明显上升(P <0.05),模型组病理损伤程度显著增加;与模型组比较各治疗组肾功能明显改善,血肌酐及血尿素氮均呈下降趋势(P <0.05),SOD明显上升及MDA明显下调(P <0.05),血清IL-6水平明显减少(P <0.05),IV-C表达明显下降,且均呈剂量依赖关系;高剂量组与科素亚组比较差异无显著性(P> 0.05)。与模型组比较各治疗组病理损伤减轻。结论:中药肾毒宁方能改善肾功能,具有一定肾保护作用,延缓肾功能损伤,其机理可能与其通过调节炎症介质,抗氧化作用,进而产生抗纤维化作用有关。
Objective: To analysis the renal protective efects of Shenduning particles (肾毒宁方) on renal failure of 5/6 nephrectomized rats with chronic renal failure. Methods: 120 SD rats were randomly divided into the sham operation group (20 rats) and the operation group (100 rats), The operation group underwent 5/6 nephrectomy. After surgery 2 weeks, the operation group was randomly divided into model group, Shenduning low, medium and high dose groups and western medicine group, 18 rats in each group. After 2 weeks, renal toxicity in each group were given saline, losartan and corresponding dose of Shenduning stomach. After 2 months of treatment, the rats were killed. The renal tissues were examined for the detection of serum superoxide dismutase and malondialdehyde and serum creatinine and blood urea in rats, 24 hour urinary protein quantitative changes. The level of IL-1β in renal tissue was detected by ELISA. Results: Compared with the model group, (P〈0.05), and the level of IL-1β was signifcantly decreased (P〈0.05), and IV-C of renal tissue in the model rats showed high expression respectively, signifcantly higher than every treatment groups (P〈0.05). Conclusion: Shenduning can improve renal function, with a certain degree of renal protective efect, delay renal injury, its mechanism may be through its regulation of infammatory mediators, and then produce antioxidant.
