重楼皂苷I对大鼠原代成骨细胞增殖的影响及机制研究

Effect of polyphyllin I on the proliferation of primary osteoblasts of rats and the possible mechanisms

目的:研究重楼皂苷I(PPL I)对大鼠原代成骨细胞增殖的影响及其机制。方法:选择酶消化法分离大鼠原代成骨细胞并进行培养,加入含不同浓度PPL I(3、10、30、100μmol/L)的培养液,采用CCK-8试剂盒、EdU试剂盒、碱性磷酸酶活性试剂盒检测大鼠原代成骨细胞的增殖情况,采用免疫印迹法观察PPL I(3、10、30μmol/L)对大鼠成骨细胞中骨保护蛋白(OPG)/核因子-κB受体活化因子配体(RANKL)表达的影响。结果:PPL I可有效地促进大鼠原代成骨细胞的增殖,降低RANKL的表达,升高OPG的表达,呈浓度依赖性。结论:PPL I可能通过影响OPG/RANKL通路,发挥促进大鼠原代成骨细胞增殖与分化的作用。

Objective：To investigate the effect of polyphyllin I on the proliferation of primary osteoblasts of rats and the possible mechanisms. Methods： Primary cultures of rats osteoblasts were obtained by enzyme digestion and administered with different concentrations of polyphyllins （3, 10, 30, and 100 μmol/L）. Then, CCK8 cell counting kit, EdU kit, and alkaline phosphatase activity detection kit were used to observe the proliferation of primary osteoblasts of rats. Western blot analysis was employed to assess the expression of osteoprotegerin （OPG） and receptor activator of nuclear factor-κ B ligand （RANKL） in primary osteoblasts treated with 10, 30, and 100 μg /ml of polyphyllins. Results： Polyphyllin I could effectively promote proliferation of primary osteoblasts and upregulate the expression of OPG and downregulate RANKL expression. Conclusions： Polyphyllins may promote primary osteoblasts of rat proliferation and differentiation through regulating OPG/RANKL signaling pathway.
