摘要
目的:观察肉毒素A(BTXA)局部注射对于兔耳瘢痕形态以及瘢痕内成纤维细胞增殖活性的影响。方法:将10只新西兰大耳白兔建立的120处兔耳瘢痕模型分为肉毒素A治疗组(T组)和对照瘢痕组(S组)各60个创面。术后28d时,收集两组瘢痕组织,分别进行HE染色、MTT法检测成纤维细胞增殖活性和电泳法测定TGF-β1表达。结果:组织学检查及瘢痕增生指数结果提示BTXA局部注射可抑制瘢痕内成纤维细胞的过度无序增殖;MTT法检测结果提示BTXA作用后的成纤维细胞活性明显较未治疗组降低(P<0.05);TGF-β1表达测定,治疗组(T组)TGF-β1条带明显小于对照组(S组),差异有统计学意义(P<0.05)。结论:通过对瘢痕中成纤维细胞增殖活性的抑制,BTXA可以降低成纤维细胞对于胶原等细胞外基质的合成以及下调细胞因子TGF-β1表达,起到抑制瘢痕增生的效果。
Objective:To observe the effects of local injection of Botulinum toxin A on the morphology of rabbit ear scar and the proliferation activity of fibroblasts . Methods:Ten New Zealand white rabbits were used to establish 120 rabbit ear scar models, Botulinum toxin A treatment group (T group) and control scar group (S group) for 60 wounds. At 28 days after operation, the scar tissue was collected , HE staining, MTT assay and Electrophoresis were used to detect the proliferation of fibroblasts and the expression of cytokine TGF-β1. Results:Histological examination and scar hyperplasia index indicated that local injection of BTXA could inhibit the excessive disordered proliferation of fibroblasts in scars. The results of MTT assay showed that the activity of fibroblasts after BTXA treatment was significantly lower than that of the untreated group ( P 〈0.05). The TGF-β1 band in the treatment group (T group) was significantly smaller than that in the control group (S group), and the difference was statistically significant ( P 〈0.05). Conclusion: By inhibiting the activity and proliferation of fibroblasts in scars, Botulinum toxin A can reduce the synthesis of fibroblasts for extracellular matrix such as collagen and down-regulate the expression of cytokine TGF-β1, thereby inhibiting scar hyperplasia.
