过表达CUL4A对肺腺癌A549细胞生物学功能的影响

The effects of overexpression of CUL4A on the biological behavior of lung adenocarcinoma A549 cells

目的:探究CUL4A对人肺腺癌细胞株A549生物学行为的影响。方法:通过慢病毒感染的方法,构建CUL4A过表达的A549细胞株,通过体外平板克隆和Transwell实验检测A549细胞株的增殖、迁移和侵袭能力。结果:通过RT-PCR和Western-blotting证实CUL4A过表达的A549细胞株构建成功。与对照组的细胞相比,上调CUL4A的表达能够促进A549细胞的增殖、迁移和侵袭能力。结论:我们的研究结果强调了CUL4A在人肺腺癌细胞株A549中的重要性,并提示CUL4A不仅可能与肺癌的发生和转移有关,而且可能成为肺癌潜在的治疗靶点以及生物标志物。

Objective： To study the effects of CUL4A on the biological behavior of the human lung adenocarcinoma cell line A549. Methods： The expression level of CUL4A was examined by RT-PCR and Western-blotting. Forced expression of CUL4A was mediated by lentiviruses. The proliferation,migration and invasion ability of A549 cells was measured by the colony-forming assay and Transwell based methods in vitro. Results： The successful transfection was confirmed by RT-PCR and Western-blotting in A549. Overexpression of CUL4A can increase the proliferation,migration and invasion ability of A549 cells. Conclusion： Our results highlight the significance of CUL4A in the human lung adenocarcinoma cell line A549 and suggest that CUL4A could not only be related to the development and metastasis of lung cancer but also be a promising therapy target and a potential biomarker in lung cancer.
