基于TCGA数据库挖掘UBE2C在肾透明细胞癌中的表达及预后意义 被引量:4

摘要 目的:通过研究泛素偶联酶E2C(ubiquitin-conjugating enzyme E2C,UBE2C)在肾透明细胞癌(kidney renal clear cell carcinoma,KIRC)组织中的表达,阐明其对KIRC早期诊断及预后的影响。方法:利用Oncomine、GEPIA数据库和人类蛋白质表达图谱(The Human Protein Atlas,HPA)分析UBE2C在KIRC组织中mRNA和蛋白质水平的变化。运用Linked Omics和GEPIA数据库阐述UBE2C表达与KIRC临床病理学参数的相关性及对预后的影响。在String-DB数据库中探索UBE2C基因在细胞信号转导通路中的位置以及与其关系密切的上下游基因。结果:UBE2C的mRNA在KIRC中表达增加,并且与KIRC病理分级、TNM分期呈正相关,且与种族和人种相关(P <0. 05)。我们应用TCGA分析的结果显示:高水平UBE2C是KIRC总生存率和无病生存率不良的独立预后因素(P <0. 01)。IHC结果证实,与正常肾组织相比,KIRC中UBE2C的蛋白表达量显著升高(P <0. 05)。与UBE2C相关的蛋白有ANAPC10、ANAPC11、ANAPC2、ANAPC4、ANAPC5、AURKA、AURKB、CCNB1等,可能参与细胞周期检查点及蛋白泛素化等细胞功能。结论:通过TCGA数据库信息挖掘发现,UBE2C在KIRC组织中呈高表达,且与患者生存期有关,将为后续的肾癌研究提供重要的理论依据。 Objective: To investigate the role of ubiquitin-conjugating enzyme E2C( UBE2C) in early diagnosis and prognostic significance of kidney renal clear cell carcinoma( KIRC) by studying the expression of UBE2C in the tissue of KIRC. Methods: The UBE2C mRNA and protein expression in KIRC tissues and normal renal tissues were measured by using Oncomine database,GEPIA and Human Protein Atlas( HPA). The correlations between UBE2C expression and prognosis on KIRC clinicopathological parameters were analyzed by Linked Omics and GEPIA. The location of the UBE2C gene in the cell signal transduction pathway and the upstream and downstream genes associated with it were explored in the String-DB database. Results: At the mRNA level,UBE2C was higher in KIRC than normal kidney tissues,and the expression level was positively correlated with the pathological stage and TNM stage of KIRC,as well as correlated with ethnicity and race( P 0. 05). The results of TCGA indicated that high UBE2C mRNA expression was an independent prognostic factor for poor overall survival and disease-free survival rate in KIRC( P 0. 01). Compared with the normal kidney tissues,immunohistochemical staining showed that UBE2C expression was significantly up-regulated in the KIRC tissues( P 0. 05). UBE2C related proteins are ANAPC10,ANAPC11,ANAPC2,ANAPC4,ANAPC5,AURKA,AURKB,CCNB1,etc,may be involved in cell cycle checkpoint,protein ubiquitination and other cellular functions. Conclusion: UBE2C was increased in KIRC and was related to the patients 'survival. It would be an important theoretical support for the further research.
