miR-145在浆液性卵巢癌中的表达及对卵巢癌SKOV3细胞增殖的影响 被引量:6

摘要 目的:检测miR-145在浆液性卵巢癌组织及卵巢癌SKOV3细胞中的表达情况,探讨其对卵巢癌SKOV3细胞增殖的影响及其可能的分子机制。方法:利用实时定量PCR技术检测43组浆液性卵巢癌患者癌及癌旁组织中miR-145的表达情况;利用实时定量PCR技术检测正常卵巢浆液腺细胞及SKOV3细胞中miR-145的表达情况;合成miR-145的模拟物miR-145 agomir,将该模拟物及对照分别转染入SKOV3细胞中,应用MTT法检测细胞增殖情况,明确miR-145对卵巢癌SKOV3细胞增殖的影响。结果:实时定量PCR结果显示,与癌旁组织相比,miR-145在浆液性卵巢癌患者癌组织中表达含量降低;与正常细胞相比,miR-145在SKOV3细胞中表达含量降低; MTT结果表明转染了miR-145 angomir的SKOV3细胞增殖受到明显的抑制。结论:miR-145在浆液性卵巢癌组织及SKOV3细胞中均表达含量降低,miR-145可以抑制卵巢癌细胞增殖,miR-145与卵巢癌患者预后密切相关,其含量增高提示好的预后,有望成为新的生物学标志物用于卵巢癌早期诊断及判断预后。 Objective: To detect the expression of miR-145 in serous ovarian carcinoma and SKOV3,and to explore its effect on the proliferation of ovarian cancer cell SKOV3 and its possible molecular mechanism. Methods: The expression of miR-145 in 43 serous ovarian carcinoma tissues and adjacent tissues was detected by real-time quantitative PCR. The real-time quantitative PCR was used to detect the expression of miR-145 in normal ovarian serous gland cells and SKOV3 cells. The miR-145 agomir was synthesized and then the miR-145 agomir and controls were transfected into SKOV3 cells respectively. The MTT assay was uesd to detect the proliferation of SKOV3 cells,exploring its effect on the proliferation of ovarian carcinoma cell SKOV3. Results: The results of real-time quantitative PCR showed that the expression of miR-145 in serous ovarian carcinoma tissues was lower than that in adjacent tissues. Compared with normal cells the expression of miR-145 was decreased in SKOV3 cells. MTT results show that the proliferation of SKOV3 cells transfected with miR-145 angomir were significantly inhibited. Conclusion: miR-145 is decreased in serous ovarian carcinoma tissues and SKOV3,miR-145 can inhibit the proliferation of ovarian cancer cells,miR-145 is closely related to the prognosis of patients with serous ovarian carcinoma,its increased expression suggests a good prognosis. miR-145 is expected to become a new biology marker for early diagnosis and prognosis.
