摘要 目的:探讨脂肪酸合成酶(fatty acid synthase,Fas)在膀胱癌患者的血清及组织中的表达及意义。方法:ELISA法测定58例膀胱癌术前术后血清和23例体检正常健康人血清中Fas蛋白的表达;免疫组织化学(SP法)测定与血清患者匹配的58例膀胱癌组织和15例前列腺增生患者的正常膀胱黏膜组织Fas的表达。结果:膀胱癌患者血清中Fas水平(34. 61±15. 83)μg/L高于健康人群血清中Fas水平(19. 14±4. 85)μg/L(P <0. 05);膀胱癌血清Fas的表达水平与肿瘤单发多发、病理分期、临床分期、淋巴转移有关(P <0. 05);膀胱癌患者组织Fas的阳性表达率为48. 7%(28/58)与健康人群组93. 3%(14/15)比较差异有统计学意义(P<0. 05);膀胱癌Fas的阳性表达与肿瘤单发多发、病理分期、临床分期、淋巴转移有关(P <0. 05)。结论:Fas蛋白在膀胱癌的发生、发展中起重要作用,膀胱癌血清中Fas水平与癌组织Fas阳性表达具有一致性。 Objective: To investigate the expression and significance of fatty acid synthase( Fas) in the serum and tissue of patients with bladder cancer. Methods: 58 patients with bladder cancer before and after operation and 23 normal were determined by ELISA method. Immunohistochemistry( SP method) was used to adetermine the expression of Fas protein in 58 cases of bladder carcinoma patients,and 15 cases of benign prostatic hyperplasia patients with normal bladder mucosa. Results: The level of Fas( 34. 61 ± 15. 83) μg/L in serum of patients with bladder cancer was higher than that in healthy population( 19. 14 ± 4. 85) μg/L( P〈0. 05). The positive expression of Fas in bladder cancer was correlated with single tumor,pathological stage,clinical stage and lymphatic metastasis( P〈0. 05).Conclusion: Fas protein plays an important role in the carcinogenesis and development of bladder cancer. The level of Fas in serum of bladder cancer is consistent with the positive expression of Fas in cancer tissue.
