阿托伐他汀调控MMP-9、Cleaved Caspase-3、Bcl-2和Bax蛋白表达对胃癌细胞增殖、周期和凋亡的影响

Effects of Atorvastatin on the proliferation,cycle and apoptosis of gastric cancer cells by regulating the expressions of MMP-9,Cleaved Caspase-3,Bcl-2 and Bax proteins

目的探讨阿托伐他汀对胃癌细胞增殖、周期和凋亡的影响及其机制。方法取对数生长期的胃癌SGC-7901细胞,加入终浓度为0、20、60和100μmol/L的阿托伐他汀,采用MTT法检测细胞增殖,流式细胞仪检测细胞周期和凋亡,Western blotting检测细胞中MMP-9、Cleaved Caspase-3、Bcl-2和Bax蛋白的表达。结果阿托伐他汀能够呈时间-浓度依赖性抑制SGC-7901细胞增殖;阿托伐他汀作用48 h后,G_0/G_1期细胞所占比例、细胞凋亡率、Cleaved Caspase-3蛋白和Bax蛋白的表达浓度依赖性升高,S期细胞比例、G_2/M期细胞比例、MMP-9蛋白和Bcl-2蛋白浓度依赖性降低。结论阿托伐他汀能够抑制胃癌细胞增殖和诱导细胞凋亡,其作用机制可能与诱导细胞G_0/G_1期阻滞、上调Cleaved Caspase-3和Bax蛋白和下调MMP-9和Bcl-2蛋白有关。

Objective To investigate the effect of Atorvastatin on the proliferation,cycle and apoptosis of gastric cancer cells and its mechanism. Methods The logarithmic growth of the gastric cancer cell line SGC-7901 was employed during logarithmic growth phase,and Atorvastatin were added at the final concentrations of 0,20,60 and100 μmol/L,respectively. The proliferation of the cells was detected by MTT assay. The cell cycle and apoptosis were measured by flowcytometry. The expressions of MMP-9,Cleaved Caspase-3,Bcl-2 and Bax proteins in the cells were detected by Western blotting. Results Atorvastatin suppressed the growth of SGC-7901 cells in a time and concentration-dependent manner. After cells treated with Atorvastatin 48 hours,the proportion of G0/G1 phase cells,the rate of apoptosis,the expressions of Cleaved Caspase-3 and Bax proteins were increased in a concentration-dependent manner.The proportion of S phase and G2/M phase cells,the expressions of MMP-9 and Bcl-2 proteins were decreased in a concentration-dependent manner. Conclusion Atorvastatin can inhibit the proliferation and induce apoptosis of gastric cancer cells,and its mechanism may be related to the induction of G0/G1 phase arrest,up regulation of Cleaved Caspase-3 and Bax proteins and down regulation of MMP-9 and Bcl-2 proteins.
