摘要
目的:初步探讨染料木素磺酸钠是否对大鼠脑缺血再灌注损伤诱导的自噬起调控作用。方法:采用MCAO动物模型和谷氨酸损伤的原代培养大鼠皮层神经元模型,给予GSS处理后,分别提取大脑皮层半暗带区组织或细胞总蛋白样本,运用Western blot方法测定样本中Beclin1、p-Beclin1和LC3-Ⅱ蛋白的表达情况。结果:与对照组相比,模型组Beclin1磷酸化水平及LC3-Ⅱ蛋白的表达均明显升高,经染料木素磺酸钠治疗后,相应表达量显著下降。结论:染料木素磺酸钠可能通过调控自噬对脑缺血再灌注损伤大鼠起神经保护作用。
Objective: To explore the protective effect of genistein sodium sulfonate (GSS) on rat cerebral ischemia/reperfusion injury(I/RI) and investigate the expression of autophagy-related protein. Methods: Rats cerebral ischemia/reperfusion model were made through embolizing the middle cerebral artery, and cortical neuron injury were induced by glutamate in vitro. After treatment with GSS, the protein expression of Beclin1, p-Beclin1 and LC3-Ⅱ in half injury brain tissue or cortical neuron were evaluated by western-blotting assay. Results: The level of p-Beclin1/Beclin1 and LC3-Ⅱ were both increased in ischemic semi-injury area in I/RI rats and cortical neuron injured by glutamate, which were decreased by GSS treatment. Conclusion: GSS has therapeutic effects on cerebral ischemia/reperfusion injury rats at least partially through regulating autophagy.
