摘要 应用PCR与原位杂交技术检测了40例原发性肺癌人乳头状瘤病毒(HPV)的感染率及与肺癌不同组织类型的关系;应用PCR-RFLP技术分析P53基因第七外显子的扩增与突变。结果显示:肺癌HPV阳性率为55%(22/40例),其中SCLC9/9例,鳞癌8/16例,腺癌5/12例阳性;P53基因第七外显子在HPV阳性的标本中有5/22例扩增,RFLP分析2/5例(SCLC、鳞癌各一例)突变。探讨了HPV感染、P53基因与肺癌的关系。 Surgical samples from 40 cases of Iung cancer were studied by in situ hybridization and polymerase chain reaction (PCR) to detect the infection rate of HPV DNA and its relationship with the histologieal type of the cauces. The 7 exon in P53 gene was analysed by PCR-RFLP. The results showed: the HPV DNA positive rate in lung cancer has heen 55 percent (22/40 cases), including SCLC (9/9 cases), Squamous cell carcinoma (8/16 cases)and 5 adenocarcinoma (5/12 cases). The 7 exon amplification was indicated in 5 of the 22 HPV DNA positive cases. RFLP analysis showed that there were two mutation samples in amplificative cases. Only one HPV DNA negative cases revealed 7 exon amplification and no mutation has heen obseved. Squamous carcinoma and SCLC often occurred in larger bronchi which were infected by HPV. HPV DNA might not only directly induce P53 gene mutation, but might also can inhibit downregulated function of wild P53 protein by the easly code proten E6 from HPV DNA products. This might play a role in the pathogenesis of lung cancer.
