羊栖菜褐藻糖胶的提取工艺优化及理化性质分析 被引量:2

摘要 为了获得经济、高效的羊栖菜褐藻糖胶提取工艺,提高其抗氧化活性,以羊栖菜褐藻糖胶的得率、提取物的1,1-二苯基-2-三硝基苯肼(DPPH)自由基清除率为考察指标,采用综合加权评分法考察提取介质、提取温度、提取时间和液料比对褐藻糖胶得率及DPPH清除率的影响,并初步描述羊栖菜褐藻糖胶的理化性质.结果显示,当提取介质为蒸馏水、液料比为20∶1、提取温度为85℃、提取时间为1 h,褐藻糖胶得率为7.37%,DPPH清除率为84.13%.基于该优化工艺制备的羊栖菜褐藻糖胶的分子量分布范围为39.5–53.0 kDa和53.0–110.2 kDa,硫酸根含量为17.1%,由岩藻糖、半乳糖、葡糖糖、甘露糖、鼠李糖和木糖构成.另外,提取介质不仅会影响褐藻糖胶的提取效率,同时也会影响自由基清除能力,因而,应采用适当的提取工艺,在提高褐藻糖胶得率的同时,保留其抗氧化活性. The comprehensively weighted grading method is adopted to inspect the extraction media, the extraction temperature, the extraction time and the effect of the liquid-solid ratio to the fucoidan yield and DPPH clearance rate in order to obtain the economic and the high effective extraction process of Sargassum fusiformis fucoidan. This method aims to upgrade the antioxidant activity by means of the yield of Sargassum fusiformis fucoidan with its extraction of 1:1 dipheyl-2-DPPH free radical scavenging rate as its investigation target and primarily describe to the physicochemical property. The result displays that the extraction medium is distilled water with the of liquid-solid ratio to raw material being 20∶1, the extraction temperature is 85℃, the extraction time is one hour, the yield of fucoidan comes to 7.37% and the DPPH scavenging rate reaches 84.13%. Sargassum fusiforme fucoidan is constituted by fucose, galactosum, glucose, mannitose, isodulcitol and xylopyranose based on molecular weight of Sargassum fusiformis fucoidan supplied in the optimized preparation technology with the distribution within the scope from 39.5 – 53.0 to 53.0 – 110.2 kDa and 17.1% of the sulphate content. Furthermore, the extraction media not only affects the extraction efficiency of the fucoidan, but also affects the free radical scavenging capability. As a result, the appropriate extraction process is applied to enhance the yield of fucoidan and retain its antioxidant activity.
