氯化乙酰胆碱对急性肺水肿小鼠肺组织Na^+,K^+-ATP酶的作用 被引量:1

The Effect of Acetylcholine Chiride on Na^+,K^+-ATPase in Lung Tissue of Mice with Acute Pulmonary Edema

摘要 目的:研究氯化乙酰胆碱(Ach)对急性肺水肿小鼠肺组织Na^+,K^+-ATP酶活性及蛋白表达的影响。方法:昆明小鼠随机分为对照组(control)、10-4mol/L氯化乙酰胆碱(Ach)组、10-5mol/L Ach组、10-6mol/L Ach组、10-6mol/L Ach+10-6mol/L阿托品组,将含有不同浓度Ach的等渗生理盐水注入到小鼠气道内,应用Na^+,K^+-ATP酶活性测定试剂盒测定各组小鼠肺组织匀浆内Na^+,K^+-ATP酶的活性,应用Western-Blot方法测定各组Na^+,K^+-ATPɑ1蛋白表达。结果:与对照组比较,10-4~10-6mol/L Ach组明显增加了Na^+,K^+-ATP酶的活性(P <0. 05),10-6mol/L阿托品阻断了10-6mol/L Ach对Na^+,K^+-ATP酶的作用(P <0. 05)。Western-Blot结果显示各组Na^+,K^+-ATPaseα1蛋白表达没有差异。结论:Ach能够增加肺组织匀浆Na^+,K^+-ATP酶的活性,阿托品能够阻断该效应,但对肺组织匀浆Na^+,K^+-ATPaseα1蛋白表达没有影响。 Objective:To explore the effect of aceulcholine chloride on Na ^+ , K^ + -ATPase activity and protein expression in lung tissue of acute pulmonary edema in mice. Methods:Kunming Mice were randomly divided into control group, 10^-4 mol/L acetylcholine chloride( Ach) group, 10^-5 mol/L Ach group, 10^- 6 mol/L Ach group and 10^-6 mol/L Ach + 10^-6 mol/L atropine group. Several different concentrations of aceulcholine Chloride were injected into mice tracheal, Na + , K + -ATPase activity was measured by Na + , K + -ATPase Activity Assay kit, and Western Blot method was used for protein expression. Results: Compared with the control group, while 104 -10^-6 mol/L acetylcholine chloride significantly increased the Na + , K + -ATPase activity (P 〈 0.05) , 10 ^-6 mol/L atropine blocked 10 6mol/L acetylcholine chloride on the effect of Na + , K + -ATPase activity (P 〈 0.05 ). But there is no significantly change in Na + , K + -ATPase α1 protein expression. Conclusion : Acetylcholine chloride can increased the Na + , K + -ATPase activity in lung tissue homogenate, and atropine abolished such effect. But there is no difference in Na + ,K + -ATPase α1 protein expression.
