高粱种子萌发期耐盐材料的筛选与鉴定 被引量:9

摘要 【目的】种子萌发期,筛选、鉴定耐盐材料对保证高粱群体数量、高粱产量以及培育高粱耐盐品种都具有重要意义。【方法】首先以粒用高粱BTx623和甜高粱Rio为代表,采用Na Cl模拟盐胁迫,设置0(CK),0. 2%、0. 4%、0. 6%、0. 8%、1. 0%、1. 2%、1. 4%、1. 6%、1. 8%和2. 0%不同浓度梯度处理,确定Na Cl初步筛选浓度。其次,利用Na Cl初步筛选浓度对全球收集的396份高粱进行初步筛选,筛选出较耐盐的材料逐渐加大选择压力,最终筛选出强耐盐材料。【结果】经过以上试验,确定了大批材料的初步筛选时Na Cl浓度为1. 2%较为适宜。396份高粱材料经过1. 2%、1. 4%、1. 6%、1. 8%逐步筛选、鉴定,最终筛选出5份耐盐能力很强的高粱材料,其中4份是中国的粒用高粱,1份是美国的粒用高粱。【结论】高粱种子萌发期,对大量种质资源进行耐盐材料筛选时,需要确定较为适宜的Na Cl筛选浓度以及需要逐级加大选择压力。 [ Objective] At the seed germination stage of sorghum, screening and identification of the salt-resistant materials, are important for assuring crop population, yield and cultivating the salt-resistant varieties. [ Method] Firstly, grain sorghum BTx623 and sweet sorghum Rio, were separately treated with0(CK), 0.2 %, 0.4 %, 0.6 %, 0.8 %, 1.0 %, 1.2 %, 1.4 %, 1.6 %, 1.8 % and 2.0 % NaCl, which was aimed at determining the initial screening concentration of NaCl. Secondly, 396 sorghum materials, from the whole world, were treated by the NaCl initial screening concentration to screen the salt-resistant materials, and then gradually increase selection pressure to screen the stronger salt-resistant materials. [ Result] Our data showed that 1.2 % NaCl was appropriate for initial screening for a large num- ber of materials. 396 sorghum materials were treated by 1.2 % , 1.4 %, 1.6 % and 1.8 % NaCl stepwise selection, 5 higher salt-resistant materials were obtained, 4 materials were from China, 1 material was from America, which were all grain sorghum. [ Conclusion ] At the seed germination stage of sorghum, we need determining the appropriate screening concentration of NaCl and gradually increasing the selec- tion pressure during screening and identification of the salt-resistant materials.
