3株传染性法氏囊病病毒的分离与鉴定

摘要 本研究于2014—2017年从山东省发生传染性法氏囊病的鸡群中分离到3株传染性法氏囊病病毒(Infectious bursal disease virus,IBDV)毒株,通过RT-PCR技术对3株分离毒株进行鉴定,并测定其ELD50以及对3周龄SPF鸡的致死率。结果如下:3株传染性法氏囊病病毒分离株均可导致70%以上SPF鸡的死亡,属于强毒株。通过特异性引物对3株分离株VP2基因的高变区进行扩增并测序,序列分析结果显示:3株分离株与5个超强毒株均在同一个大的分支上,同源性在96. 3%~99. 3%之间;与疫苗株B87和D78分属于两个分支,同源性仅为88. 7%~90. 6%。本研究为山东地区传染性法氏囊病的防治提供了有益参考。 Three infectious bursal disease virus (IBDVs) isolates were isolated from diseased chickens in Shandong Province in 2014-2017. These virus strains were detected by RT-PCR and inoculated into 3-week-old SPF chickens to determine their ELD 50 and pathogenecity. The results showed that the mortality of the infected SPF chickens was equal to or greater than 70.0%,which indicated these isolates belonged to vvIBDV. The specific primers were used to amplify and sequence the high-varying regions of the VP2 gene of the three strains.The sequence analysis results showed that the three isolates were in the same big branch with other five vvIBDVs,and the homology was 96.3%~99.3%;the three isolates were classified into different branch from vaccine strains B87 and D78, and the homology was only 88.7%~90.6%. This study provided a useful reference for the prevention and treatment of IBDV in Shandong Province.
