摘要
选用SD雄性大鼠,分为安静对照组(QC组)、运动对照组(EC组)和运动加α-硫辛酸组(EPL组),每组8只。第6周最后1天取股四头肌,制备线粒体样品,分别测试大鼠股四头肌线粒体抗氧化指标、表征线粒体功能的标志酶活性,探讨线粒体营养素α-硫辛酸(ɑ-LA)对大鼠股四头肌线粒体抗氧化能力与能量代谢标志酶活性的影响。结果显示,EC组、EPL组抗氧化酶活性与QC组比较下降(P<0.01或P<0.05);与EC组比较,EPL组抗氧化酶活性上升(P<0.05或P<0.01)。与QC组比较,EC组、EPL组MDA和H2O2含量升高(P<0.01);与EC组比较,EPL组MDA和H_2O_2的含量下降(P<0.01)。与QC比较,EC组和EPL组三种ATPase活性下降(P<0.01或P<0.05);与EC组比较,EPL组三种ATPase活性上升(P<0.05或P<0.01)。与QC组比较,EC组功能标志酶活性下降(P<0.05或P<0.01),EPL组功能标志酶活性下降(P<0.05);与EC组比较,EPL组功能标志酶活性上升(P<0.05或P<0.01)。EPL组大鼠运动至疲劳的时间延长30.05%。结果表明,α-LA对运动训练大鼠体重增长无不良影响,对线粒体的结构与功能具有维护作用,可保证线粒体的氧化供能效率,提高大鼠的有氧运动水平,具有较强的抗疲劳功效。
To investigate the effects of mitochondrial nutrient α-lipoic acid on mitochondrial antioxidant capacity and energy metabolism marker enzyme activity in rat quadriceps muscle. SD rats were divided into quiet control group (QC group), exercise control group (EC group) and exercise plus α-lipoic acid group (EPL group), with 8 rats in each group. On the last day of the sixth week, the quadriceps muscle was taken and mitochondrial samples were prepared to test the mitochondrial antioxidant index and the marker enzyme activity of mitochondrial function in the rat quadriceps muscle. The results showed that the activities of antioxidant enzymes in EC group and EPL group were lower than those in QC group (P〈0.01 or P〈0.05). Compared with EC group, the activity of antioxidant enzymes in EPL group increased (P%0.05 or P〈0.01). Compared with QC group, MDA and H2O2 levels in EC group and EPL group were increased (P〈0.01). Compared with EC group, MDA and H2 02 levels in EPL group were decreased (P〈0.01). Compared with QC, the three ATPase activities in the EC group and the EPL group decreased (P〈0.01 or P%0.05). Compared with the EC group, the three ATPase activities in the EPL group increased (P〈0.05 or P〈0.01). Compared with the QC group, the activity of the functional marker in the EC group decreased (P〈0.05 or P〈0.01), and the activity of the functional marker in the EPL group decreased (P〈0.05). Compared with the EC group, the activity of the functional marker in the EPL group increased (P〈0.05 or P〈0.01). The time from exercise to fatigue in the EPL group was prolonged by 30.05 %. The results showed that α-LA had no adverse effects on the weight gain of exercise-trained rats. The structure, function and mainte nance of mitochondria ensured the oxidative energy supply efficiency of mitochondria and increased the aerobic exercise level of rats,and has a strong effect of anti-fatigue.
