缺血预处理对肝缺血再灌注后氧自由基损伤的保护作用

Protective Effect of Ischemic Preconditioning on Liver Injury from Oxygen Free Radicals after Hepatic Ischemia-reperfusion

目的 探讨缺血预处理(PC)对肝缺血再灌注损伤的保护作用机制。方法 将18只犬随机分为三组:非缺血对照组、缺血再灌注组和缺血预处理组。对各组肝上下腔静脉血进行谷丙转氨酶(ALT)、谷草转氨酶(AST)、乳酸脱氢酶(LDH)以及丙二醛(MDA)和超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化酶(GSH-PX)活性及总抗氧化(TAX)能力测定。结果 全肝缺血再灌注后ALT、AST、LDH和MDA含量明显上升(P<0.01),SOD、CAT、GSH-PX活性及TAX能力明显下降(P<0.01,P<0.05);而缺血预处理组与缺血再灌注组比较,ALT、AST、LDH和MDA含量明显下降(P<0.01,P<0.05),SOD、CAT、GSH-PX活性及TAX能力明显上升(P<0.01,P<0.05)。结论 缺血预处理能增强肝组织自身抗氧化能力,减轻肝缺血灌注后氧自由基对肝脏的损伤。

To study the mechanisms of protective effect of ischemic preconditioning(PC) on hepatic ischemia-reperfusion(I- R)injury.Metheds Eighteen dogs were randomly divided into three groups;nonischemic controls(C) group( n = 6)、ischemia-reperfusion group(I- R)( n =6) and ischemic preconditioning( PC) group(MHTT)( n =6). The content of the marker enzymes of liver damage (ALT, AST, LDH) and malondialdehyde(MDA) and activities of superoxide dismutase(SOD),catalase (CAT),glutathione peroxidase(GSH-PX),total antioxidase(TAX) in inferior vena cava blood above liver were measured in each group. Results After I - R the content of ALT、AST、LDH and MDA were significantly elevated( P &lt; 0.01), SOD, CAT, GSH -PX,ACT activeties were declined obviously( P &lt; 0.01, P &lt; 0.05),but the content of ALT,AST,LDH and MDA were obviously lowere in PC group than that in I - R group ( P &lt;0.01, P &lt; 0.05); and the content of SOD, CAT, GSH-PX, ACT activeteis were obviously higher than that in I - R group( P &lt; 0.01, P &lt; 0.05). Conclusion Ischemia-reperfusion may enhance liver ability of oxidation-resistance,and reduce the oxygen free radical injury to liver after ischemia-reperfusion.
