β-甲基戊二酸单甲酯的合成

Synthesis of Monomethyl β-Methylglutarate

摘要 本研究创新性地开发了一条合成β-甲基戊二酸单甲酯的新路线。β-甲基戊二酸与甲醇酯化得β-甲基戊二酸双甲酯(3),再利用氢氧化钡对酯的温和碱解反应,借助双官能团相互作用的电子效应,促使3中只有一个酯基官能团参与碱解反应,高选择性地生成β-甲基戊二酸单甲酯钡盐,然后经酸化、萃取、精馏得目标产物,总收率90.3%。此工艺已经过中试验证。 A novel synthetic process for monomethyl β-methylglutarate was developed creatively. Dimethyl β-methyl pentanedioate (3) was obtained by esterification of β-methylglutaric acid with methanol. Then, in the presence of barium hydroxide, only one ester group in 3 was hydrolyzed to produce barium monomethy1β-methylglutarate with high selectivity. Then the latter was subjected to acidification, extraction and rectification to afford the target product with a total yield of 90.3 %. This process had been validated in pilot scale.
