摘要
乙酰胆碱酯酶抑制-分光光度法是目前监管中应用最为广泛的快速筛查蔬菜中有机磷和氨基甲酸酯类农药残留的快速检测方法。对该方法进行了多项条件的优化,结果表明,酶活性ΔA0控制在0.2~0.3时,抑制率能真实反映样品阳性结果;孵育温度是影响结果精密度的重要因素,部分仪器自带恒温装置的温控稳定性较差,比色槽温度变化较大,导致反应结果的平行性和重现性较差,试样在恒温水浴锅中的反应结果重现性很好;配制标准溶液时,应将丙酮含量控制在≤1%时,丙酮对试验的影响可以忽略。
Acetylcholinesterase-spectrophotometry is the most widely used method for rapid detection of organophosphorus and carbamate pesticide residues in vegetables. However,in practical application,the method is seriously limited by nmnerous peripheral influencing factors and low accuracy. The optimization of Acetylcholinesterase-Spectrophotometry for determination of organophosphate and earbamate pesticide residues in vegetables were studied in this paper.Controlling the AA value between 0.2-0.3 and its results were given,which could improve the accuracy of positive test results. Extra thermostatie bath was chosen instead of device came with instrmnent and the results showed greatly improved reproducibility and parallelism. In order to avoid the influence of acetone on enzyme activity,the added amount of acetone should be controlled less than 1% when preparing standard solution.
