大叶贯众多酚对免疫抑制小鼠免疫功能和抗氧化的影响 被引量:4

Effects of large phenols on immune function and anti-oxidation in immunosuppressed mice

摘要 为测定大叶贯众多酚(CMPE)对环磷酰胺(CY)诱导的免疫抑制小鼠模型免疫功能和抗氧化功能的影响,笔者将90只雄性BALB/c小鼠随机分为6组,除健康对照组和阳性对照组外,3个试验组和模型组均腹腔注射80 mg/(kg·d)环磷酰胺,连续3 d,然后3个试验组灌胃给予50、100、200 mg/kg的CMPE,健康对照组和模型对照组给予生理盐水,阳性对照组灌胃给予400 mg/kg的CMPE,持续18 d。于最后一次给药后第24小时,每组抽取3只小鼠用碳粒廓清试验检测巨噬细胞的吞噬指数,3只小鼠用MTT法测定淋巴细胞增殖,其余小鼠称重后颈椎脱臼处死,采集内脏并称重,计算脾、胸腺指数,测定心、肝、肾匀浆液中T-AOC、MAD、CAT、SOD和GSH-Px水平。结果显示,3个CMPE组以剂量依赖的方式增加小鼠的脾指数、胸腺指数、脾淋巴细胞增殖和巨噬细胞的吞噬指数、SOD、CAT、GSH-Px和TAOC活性;而3个CMPE组MDA水平以剂量依赖的方式降低。结果表明,CMPE可以逆转环磷酰胺诱导的免疫器官萎缩,增强免疫抑制小鼠的细胞免疫和巨噬细胞功能,有效清除自由基,可作为潜在的免疫调节剂。 90 male BALB/c mice were randomly divided into six groups including 3 CMPE experimental groups,1 healthy control group,1 positive control group and 1 model control group.The mice in the three CMPE groups and the model control group were given cyclophosphamide at 80 mg/(kg·d) via intraperitoneal injection for 3 days.Three CMPE groups were administrated at 50,100 or 200 mg/kg CMPE according to body weight via gavage.The healthy control group and the model control group were given physiological saline,and the positive control group was orally administered at 200 mg/kg CMPE,lasting 18 days.At 24th hour after the last administration,3 mice of each group was detected in macrophage phagocytic index by carbon clearance test,and 3 mice was measured in lymphocyte proliferation by MTT method.The rest of the mice were put to death by dislocated cervical vertebra after weighing,their internal organs were collected and weighed,spleen and thymus indexes were calculated,and the levels of proteins,T-AOC,MAD,CAT,SOD and GSH-Px in the heart,liver and kidney homogenate were measured.In result,the 3 CMPE groups in a dose-dependent manner increased spleen index,thymus index,spleen lymphocyte proliferation and macrophage phagocytic index,and the phagocytic activity of the high dose group returned to normal level.The above-mentioned results indicated that CMPE could reverse the immune organ atrophy induced by cyclophosphamide immunosuppressed mice,and enhance the cellular immunity and macrophage function.
