摘要
目的优选延胡索药材的最佳闪式提取工艺。方法采用闪式提取技术,以脱氢紫堇碱的得率为指标,采用反相高效液相色谱法测定。选择乙醇浓度、浸泡时间、料液比、提取时间为考察因素,选用L9(34)正交设计-重复试验法优选延胡索药材中脱氢紫堇碱(DHC)最佳闪式提取工艺。结果延胡索药材的DHC最佳闪式提取工艺条件为提取电压70V,乙醇浓度体积分数50%,浸泡时间20 min,提取时间2 min,料液比1∶20。结论闪式提取延胡索药材效率高、时间短,可成为提取脱氢紫堇碱的快速有效方法。
Objective To optimize the flash extraction process for dehydrocorydaline isolated from Corydalis Rhizoma. Methods We used the flash extraction process to extract dehydrocorydaline from Corydalis Rhizoma and performed HPLC with its yield as the observation indexes. Selecting ethanol concentration, soaking time, ratio of liquor to material and extraction time as the examination factors, we adopted L9(34) orthogonal design-duplicate test to select the optimum flash extraction process. Result The optimum flash extraction process conditions were as follows: extracting voltage at 70 V, ethanol at the volume fraction of 75%, soaking for 20 min, extracting for 2 rain, and solid to liquid ratio being 1 : 20. Conclusion The flash extraction process for Corydalis Rhizoma medicinal materials has the advantage of higher yield and shorter time, be used for extracting dehydrocorydaline from Corydalis Rhizoma.
