人肺巨细胞癌高转移株分化的形态学、免疫组化及癌基因表达的实验研究

摘要 本文应用维甲酸(RA)对肺巨细胞癌高转移株(PLA-801-D)进行了诱导分化研究,结果显示:诱导后PLA-801-D细胞出现成熟分化的特征。细胞生长曲线下降,分裂指数减少,倍增时间延长;细胞体积增大,呈铺砖样排列。电镜下,细胞器及中丝增多,找到张力原纤维,细胞连接增多等成熟分化特征。免疫组化染色,细胞角蛋白与波形蛋白均为阳性,说明肿瘤细胞可能是较幼稚的上皮细胞。癌基因DNA-mRNA斑点杂交显示:诱导后的细胞v-src墓因表达增加,N-ras基因表达下降。证实这两种基因在肿瘤的恶性转化中可能起一定作用。 Differentiation of high metastatic strain from lung giant cell carcinoma cell line (PLA-801-D) was induced by retinoic acid(RA). The result showed that induced PLA-801-D cells seemed to have some features toward mature differentiation such as delayed cell doubling time, the cells being larger, more organnella in cytoplasm and more junctions between cells. Immunohistochemical staining for cytokeratin and vimentin was positive in induced cells. Oncogenes expression of v-src increased while that of N-ras decreased. These results denoted that v-src and N-ras might both play an important role in malignant transformation.
