毛花苷C促进肝癌细胞Huh-7凋亡并增强caspase-7的活化

Lanatoside C promotes apoptosis and increases activation of caspase-7 in Huh-7 cells

目的研究毛花苷C对肝癌Huh-7细胞增殖的影响及其对caspase-7的活化作用。方法以不同浓度的毛花苷C作用于Huh-7细胞,通过细胞增殖抑制实验及克隆形成实验,检测毛花苷C对Huh-7细胞增殖的抑制作用;采用流式细胞仪检测其对Huh-7细胞凋亡的影响,并用激光共聚焦显微镜采集细胞凋亡染色图像;通过Western blot分析细胞凋亡蛋白caspase-7的活化水平。结果毛花苷C能明显抑制Huh-7细胞的增殖,实验组与对照组间的差异具有统计学意义(P<0.05),并呈量效依赖关系;通过流式细胞仪与激光共聚焦显微镜,观察到毛花苷C诱导凋亡细胞数增加;Western blot检测结果显示,毛花苷C可增强凋亡相关酶caspase-7的活化水平。结论毛花苷C明显促进肝癌Huh-7细胞凋亡并增强凋亡相关酶caspase-7的活化水平。

Aim To investigate the effects of lanatoside C on the proliferation of hepatocellular Huh-7 cells and the activation of caspase-7. Methods The Huh-7 cells were treated with different concentrations of lanatoside C, and the proliferation of Huh-7 cells was determined by cell proliferation assay and clone formation assay. The apoptosis was determined by flow cytometry. Apoptotic staining images were collected by laser scanning confocal microscopy. And the activation level of caspase-7 was analyzed by Western blot. Results The proliferation of Huh-7 cells was significantly inhibited by lanatoside C, and there was a significant difference between the lanatoside C groups and the control group in a dose-dependent manner（ P 〈0.05）. The number of apoptotic cells was higher than that of control group observed by flow cytometry and laser confocal microscopy. Western blot assay indicated that lanatoside C enhanced the activation of caspase-7 in Huh-7 cells. Conclusions Lanatoside C significantly promotes the apoptosis of hepatocellular Huh-7 cells and enhances the activation of caspase-7.
