摘要
目的:采用HPLC对葛根饮片、水煎剂和配方颗粒的指纹图谱进行相关性研究,考察饮片、水煎剂与配方颗粒学成分的变化。方法:选用XAqua C18柱(250 mm×4. 6 mm,5μm),以甲醇-0. 1%冰醋酸水溶液为流动相进行梯度洗脱,流速为1 m L·min-1,柱温为25℃,检测波长为250 nm,采集时间为40 min。结果:分别建立10批葛根饮片、10批葛根水煎剂与10批葛根配方颗粒的指纹图谱。10批葛根配方颗粒HPLC指纹图谱的8个指纹峰均可在饮片中得到追踪,相似度均大于0. 99,并从8个共有峰中指认出葛根素。结论:葛根饮片、水煎剂和配方颗粒的主要化学成分基本相同,为配方颗粒临床提供数据支持。
Objective : To study the correlation of characteristic spectra of Puerariae Lobatae Radix decoction pieces, water decoction and formula granules by HPLC, and investigate the ilffluence of different existence forms on the main chemical constituents. Methods: HPLC with XAqua C18 column (250 mm×4. 6 ram, 5μm) was used with acetonitrile4). 1% glacial acetic acid (gradient elution) as a mobile phase, detection wavelength was at 250 nm, flow rate was 1.0 mL· min-1, and column temperature was set at 25℃. The acquisition time was 40rain. Results : The detection of 10 batches of Puerariae loba- tae radix formula granules, 10 hatches of decoction pieces and 10 hatches of water decoction were established respective- ly. Eight characteristic peaks in the HPLC characteristic spectra from 10 batches of formula granules could be tracked in the decoction pieces and water decoction, moreover, the similarity was more than 0. 99. In the eight common peaks, puerarin was verified. Conclusion: The main chemical components of Puerariae lobatae radix formula granules, water decoction and decoc- tion pieces are basically the same, and the common component contents have similar proportion. The method could be used for the quality control of Puerariae lobatae radix formula granules.
