摘要
目的探讨膀胱癌组织中ZO-1相关性核酸结合蛋白(ZONAB)的表达量与膀胱癌恶性程度的相关性。方法纳入2015年6月至2017年10月在我院泌尿外科接受手术治疗的原发性膀胱癌患者78例,术中留取膀胱癌组织、癌旁正常组织各78份分别作为膀胱癌组织组、癌旁组织组。采用实时荧光定量PCR法检测标本中目标基因表达量,对比两组中ZONAB基因及增殖、凋亡、侵袭基因信使RNA(mRNA)表达量的差异,Pearson检验分析膀胱癌组织中ZONAB m RNA表达量与上述增殖、凋亡、侵袭基因mRNA表达量的关系。结果膀胱癌组织组ZONAB mRNA的表达量高于癌旁组织组(P <0.05);增殖基因(KPNA2、YAP、HSG)、凋亡抑制基因(Bcl-2、Livin、Survivin)和侵袭基因(SNCG、TRPM 8、PARP-1)mRNA的表达量高于癌旁组织组,凋亡基因(Caspase-3、Caspase-7)mRNA的表达量低于癌旁组织组(P <0.05)。Pearson检验发现,膀胱癌组织中ZONAB mRNA表达量与增殖基因(KPNA2、YAP、HSG)、凋亡抑制基因(Bcl-2、Livin、Survivin)、侵袭基因(SNCG、TRPM 8、PARP-1)mRNA的表达量呈正相关,与凋亡基因(Caspase-3、Caspase-7)mRNA的表达量呈负相关(P <0.05)。结论膀胱组织中ZONAB表达量增高,其与癌细胞的增殖、凋亡、侵袭活性相关,可作为膀胱癌辅助诊断及恶性程度判断的可靠指标。
Objective To study the expression of ZO-1 associated nucleic acid binding protein(ZONAB) in bladder cancer and its correlation with the indication of malignancy. Methods A total of 78 cases of bladder cancer patients underwent surgical treatment in our hospital during June 2015 to October 2017, were selected as research subject. Bladder cancer tissue and normal tissue specimens were used as bladder cancer tissue group and paracancerous tissue group respectively. The target gene expression was detected by real-time quantitative PCR, and the difference of ZONAB, proliferation, apoptosis and invasion genes mRNA expression between two groups was compared. Further, Pearson test was used to examine the relationship of the expression of ZONAB m RNA in bladder cancer tissue with the expression of above proliferation, apoptosis and invasion genes mRNA. Results The expression of ZONAB mRNA in bladder cancer tissue group was higher than that in paracancerous tissue group(P〈0.05); the expressions of proliferating genes(KPNA2, YAP, HSG), apoptosis inhibiting genes(Bcl-2, Livin, Survivin), and invasion genes(SNCG, TRPM 8, PARP-1) mRNA in bladder cancer tissue group were higher than those in paracancerous tissue group; the expression of apoptosis genes(Caspase-3, Caspase-7) mRNA in bladder cancer tissue group was lower than that in paracancerous tissue group(〈P0.05). Pearson test showed that the expression of ZONAB mRNA in bladder cancer tissue was positively correlated with the expression of proliferation genes(KPNA2, YAP, HSG), apoptosis inhibiting genes(Bcl-2, Livin, Survivin), and invasion genes(SNCG, TRPM 8, PARP-1) mRNA, and negatively correlated with the expression of apoptosis genes(Caspase-3, Caspase-7) mRNA(P〈0.05). Conclusion The increased expression of ZONAB in bladder tissues is associated with the proliferation, apoptosis and invasion of bladder cancer cells. It can be used as a reliable marker for the adjuvant diagnosis and the malignant indication of bladder cancer.
