摘要 热休克蛋白47(HSP47)是一种促进胶原分泌的特异性分子伴侣,其与多种人类和实验性动物肾纤维化疾病密切相关,抑制其表达可减少胶原分泌,进而延缓纤维化疾病发展,以其为靶点的治疗为临床治疗肾脏纤维化提供一个新的治疗方法。本文就HSP47在肾纤维化疾病中的作用及以其为靶点治疗做一综述。 Heat shock protein 47(HSP47) is a collage-specific molecular chaperone that is required for molecular maturation of various types of collagens,it has a closed association between increased expression of HSP47 and excessive accumulation of collages in scar tissue of various human and experimental renal fibrotic diseases,and the suppression of HSP47 expression can reduce accumulation of collagens to delay the progression of fibrotic renal diseases.It provides a new therapeutic target to manipulate collagen production that might have enormous clinical impact in controlling a wide range of renal fibrotic diseases.Here,we outline the fibrogenic role of HSP47 and discuss the potential usefulness of HSP47 as an anti-fibrotic therapeutic target.
