摘要
以珍贵树种红豆树为试材,采用形态学鉴定和分子生物学技术相结合的方法,对野生红豆树根内内生真菌进行了分离、培养、鉴定,以期揭示红豆树根围土著DSE的种类及生态学功能。结果表明:从野生红豆树根内分离得到5株疑似DSE菌株,将疑似菌株回接到红豆树幼苗,其中2种疑似菌株能够在幼苗根部形成典型的DSE侵染结构;经rDNA ITS序列分析初步确定为Acrocaly mmavagum和Leptosphaeria sp.AS7-1;2种菌株与红豆树幼苗共生后苗高较未接种幼苗(CK)分别增加49.48%和24.80%,干质量较对照增加63.63%和36.36%。可见,DSE不仅能与红豆树共生还对其生长具有一定促进作用。
Seedlings of Ormosia hosiei was taken as test material,endophytic fungi was isolated from roots to investigate dark septate endophyte.The results showed that five strains of endophytic fungi were obstained using tissue isolation method and morphological characteristics.A pot culture experiment was showed that two strains colonized and formed dark septate hyphae and microsclerotia.These results confimed that two strains were dark septate endophytes fungi.Two strains were identified as Acrocalymma vagum and Leptosphaeria sp.AS7-1 based on rDNA-ITS sequence analysis.Co-culture experiment showed that two strains had prominent effects on promoting plantlet height which were increased by 49.48% and 24.80%.Simultaneously,dry weight increased by 63.63% and 36.36% at significant level compared with the control.Dark septate endophyte had signficant promoting effects of growth.
