摘要
初步探讨雨生红球藻破壁孢子粉对Ⅱ型糖尿病大鼠的血糖及血脂的干预作用及其机理。方法:将大鼠随机分为正常组、模型组、雨生红球藻破壁孢子粉低、中、高3个剂量组、降糖药物(盐酸二甲双胍)组。除正常组外,其它各组均用高脂高糖饲料喂养4周,腹腔注射链脲佐菌素(STZ)35 mg/kg bw后继续喂养4周进行造模。造模成功后3个剂量组在给予高脂高糖饲料的同时,灌胃给予不同剂量(50,100,200 mg/kg bw)的雨生红球藻破壁孢子粉。降糖药物组灌胃给予二甲双胍(28.5 mg/kg bw)。模型组灌胃给予蒸馏水。4周后处死大鼠取血清测定血糖、胰岛素、HDL-C、LDL-C、TG、SOD、GSH-Px的活性和MDA、IL-1β、TNF-α水平。研究结果表明,雨生红球藻破壁孢子粉可显著减少糖尿病大鼠的摄食量和饮水量,显著降低血糖和胰岛素水平,显著提高血清SOD、GSH-Px活性,降低MDA、IL-1β、TNF-α水平,显著降低糖尿病大鼠LDL和TG含量,提高HDL含量。结论:雨生红球藻破壁孢子粉可显著改善糖尿病大鼠血糖及胰岛素水平,有效调节血糖和血脂紊乱。增强机体的抗氧化能力,降低炎性因子水平可能是雨生红球藻破壁孢子粉干预血糖血脂的途径之一。
To preliminary study on the intervention of the Haematococcus pluvialis to the blood sugar and lipid of type 2 diabetes rat. Method: Healthy adult male rats were placed in an environment of free drinking water for 3 days,weighing number, we randomly divided the experimental samples, into six groups: the control group, the model group,also hypoglycemic group(Metformin hydrochloride) and Haematococcus pluvialis low, medium and high dose groups. All groups, except control group, had been modeled by injecting Streptomyces(STZ) hormone with the dose of 35 mg/kg bw after having been fed with high fat diet for 4 weeks, then, to keep feeding 4 weeks. Then, three extract-fed groups were fed with high fat foods and different dose of Haematococcus pluvialis, respectively at 50, 100, 200 mg/kg bw. For the hypoglycemic group were fed with high fat diet likewise, yet with metformin intervention simultaneously(28.5 mg/kg bw). With Haematococcus pluvialis for 4 weeks lavage alloxan diabete rat, the blood samples of mice were collected,blood glucose, insulin, HDL-C, LDL-C, TG, SOD, GSH-Px activity and MDA, IL-1β, TNF-α level in the blood were measured. The results showed that Haematococcus pluvialis can significantly inhibit diabete rat weight of reduction and return to normal, reduce water intake and diet intake, reduce high blood glucose levels of diabete mice, improve the ability of antioxidation and lower TC, as well as TG, upper HDL.
