基质细胞衍生因子-1、趋化因子受体-4对子宫内膜异位症患者子宫内膜组织MMP-9和VEGF表达的影响 被引量:6

Effects of stromal cell derived factor-1 and chemokine receptor-4 on the expression of MMP-9 and VEGF in endometrium in patients with endometriosis

摘要 目的分析基质细胞衍生因子-1、趋化因子受体-4对子宫内膜异位症(EMs)患者子宫内膜组织基质金属蛋白酶-9(MMP-9)和血管内皮生长因子(VEGF)表达的影响。方法选取南通大学附属海安医院40例EMs患者,取其40份在位内膜组织标本(A组)与40份异位内膜组织标本(B组),并选择同期40例正常子宫内膜组织标本(C组)。通过免疫组织化学法与RT-PCR技术对基质细胞衍生因子-1、趋化因子受体-4、MMP-9及VEGF的表达情况进行检测。结果基质细胞衍生因子-1、趋化因子受体-4、MMP-9及VEGF在3组标本中腺上皮细胞和间质细胞中均存在表达,且主要表达于腺上皮细胞。基质细胞衍生因子-1、趋化因子受体-4、MMP-9及VEGF平均光密度值在B组中最高,其次是A组,而C组最低;3组组间比较,差异均有统计学意义(均P<0.05),且基质细胞衍生因子-1 mRNA、趋化因子受体-4 mRNA、MMP-9 mRNA及VEGF mR-NA表达量在B组中最高,其次是A组,而C组最低;3组组间比较,差异均有统计学意义(均P<0.05)。结论基质细胞衍生因子-1、趋化因子受体-4、MMP-9及VEGF过高表达与EMs疾病发生、发展密切相关,提示基质细胞衍生因子-1、趋化因子受体-4过高表达可能利用其他相关通路对MMP-9、VEGF造成影响,进而能够在EMs侵袭与血管的形成中发挥着重要的作用,故此基质细胞衍生因子-1、趋化因子受体-4过高表达可能是诊治EMs患者的重要靶点。 Objective To analyze the effects of stromal cell derived factor-1 and chemokine receptor-4 on the expression of matrix metalloproteinase-9 (MMP-9) and vascular endothelial growth factor (VEGF) in endometrium in patients with endometriosis (EMs). Methods 40 cases of EMs were selected in Nantong University Affiliated Hai'an Hospital, divided into the 40 samples of eutopie endome- trial tissue specimens (group A) and 40 samples of ectopic endometrial tissue specimens (group B) , and selected 40 cases of normal en- dometrial tissues as group C. The expression of stromal cell derived factor- 1, chemokine receptor-4, MMP-9 and VEGF were detected by immunohistochemieal method and RT-PCR technique. Results The expression of stromal cells derived factor-1, chemokine receptor-4, MMP-9 and VEGF existed on glandular epithelial cells and stromal cells in the three groups, which mainly expressed in glandular epithelial cells. The average optical density of stromal cell derived factor- 1, chemokine receptor-4, MMP-9 and VEGF were the highest in group B, followed by group A, and those levels in the group C were the lowest, which showed statistically significant differences in comparison of the three groups (all P〈0. 05) . The expression levels of stromal cell derived factor-1 mRNA, chemokine receptor-4 mRNA, MMP-9 mRNA and VEGF mRNA were the highest in group B, followed by group A and those levels in the group C were the lowest, which showed statisti- cally significant differences in comparison of the three groups ( all P〈0. 05 ) . Conclusion The high expression of stromal cell-derived fac- tor-l, ehemokine receptor-4, MMP-9 and VEGF are closely related to the occurrence and development of EMs, suggesting that the high expression of stromal cell-derived factor-1 and chemokine receptor-4 may have impact on MMP-9 and VEGF by using other related path- ways, which can play an important role in the formation of EMs and vascular invasion, so the overexpression of stromal cell derived factor- 1 and chemokine receptor-4 may be used as important targets for the diagnosis and treatment in patients with EMs.
