加味七方胃痛颗粒对裸鼠原位移植胃癌Fas、Fadd、Caspase-8、Caspase-3因子的影响

Effect of Modified Jiawei Qifang Weitong Granule on Fas,Fadd,Caspase-8 and Caspase-3 in Nude Mice with Orthotopically Transplanted Gastric Cancer

目的：探讨加味七方胃痛颗粒对裸鼠原位移植胃癌Fas、Fadd、Caspase-8、Caspase-3因子的影响及其可能的作用机制。方法：胃癌原位移植模型裸鼠84只,随机分为空白对照组、5-FU组、中药组、5-FU＋中药组各15只,剩余备用,分别进行干预,所有组于干预初始、干预1周、干预2周及干预结束时进行胃癌组织标本采集,运用RT-PCR、Western Blot技术检测胃癌组织中Fas、Fadd、Caspase-8、Caspase-3 mRNA与蛋白的表达。结果：干预1周、干预2周后,5-FU组、5-FU＋中药组、中药组胃癌组织中Fas、Fadd、Caspase-8、Caspase-3 mRNA蛋白的表达均较空白对照组有所升高,且随着时间增长,各干预组原位移植的胃癌组织中Fas、Fadd、Caspase-8、Caspase-3 mRNA蛋白的表达总体呈上升趋势,差异有统计学意义（P〈0.05）。结论：加味七方胃痛颗粒防治胃癌的机制可能是通过调节Fas、Fadd、Caspase-8、Caspase-3因子的表达,从而发挥其对肿瘤细胞的防治作用。

Objective：To investigate the effect of modified Jiawei Qifang Weitong granule on Fas,Fas-associated death domain-containing protein（Fadd）,caspase-8 and caspase-3 in nude mice with orthotopically transplanted gastric cancer and its possible mechanism. Methods：Eighty-four nude mice with orthotopically tran planted gastric cancer were randomly divided into control group,5-FU group,Chinese herbal medicinals group,and 5-FU＋Chinese herbal medicinals group,respectively. Leave the rest in reserve. All the mice were given intervention. At the initial intervention,intervention for 1 week,for 2 weeks,and the end of intervention,the specimens of gastric cancer were collected. The expressions of Fas,Fadd,caspase-8 and caspase-3 mRNAs and proteins in gastric cancer tissues were detected by reverse transcription-polymerase chain reaction （RT-PCR） and western blot. Results：After 1 week of intervention and 2 weeks of intervention,on the expression of Fas,Fadd,caspase-8 and caspase-3 mRNAs in gastric cancer tissues,5-FU group,5-FU＋Chinese herbal medicinals group and Chinese herbal medicinals group increased more than the control group. And with the increase of time,the expression of mRNAs and proteins of Fas,Fadd,caspase-8 and caspase-3 in orthotopically transplanted gastric cancer tissues increased with time （P〈 0.05）. Conclusion ： The mechanism of modified Jiawei Qifang Weitong granule in the prevention and treatment of gastric cancer may be through the regulation of the expression of Fas, Fadd,caspase-8 and caspase-3 to exert its prevention and treatment effect on tumor cells.
