利用TCGA数据集分析PPFIA3在三阴性乳腺癌中的表达及临床意义

摘要 目的初步探讨PPFIA3在三阴性乳腺癌患者中的表达及临床意义。方法从癌症基因组图谱(TCGA)数据库下载三阴性乳腺癌患者基因表达及临床特征数据,分析PPFIA3表达与患者临床特征的关系及其对预后的影响。通过构建与PPFIA3相关的共表达网络,对共表达基因集进行KEGG信号通路富集分析。结果 PPFIA3在肿瘤组织中的表达水平明显高于癌旁正常组织(P<0.01);T3~4期、淋巴结转移患者的PPFIA3表达水平均高于T1~2期、淋巴结未转移患者(P=0.025、0.027)。生存分析表明PPFIA3高表达的患者中位生存期明显短于PPFIA3低表达的患者(P=0.009)。KEGG信号通路富集分析显示与PPFIA3共表达的基因参与肿瘤坏死因子(TNF)、丝裂素活化蛋白激酶(MAPK)和雷帕霉素靶蛋白(MTOR)信号通路,氨基糖与核苷酸糖代谢等通路的信号转导。结论 PPFIA3可能通过TNF、MAPK、MTOR等信号通路参与三阴性乳腺癌细胞的增殖、转移等过程,在三阴性乳腺癌中,PPFIA3高表达提示预后不良,其可能作为预测三阴性乳腺癌患者预后的有效生物标志物。 Objective To investigate the expression and clinical significance of PPFIA3 in the triple negative breast cancer (TNBC).Method The PPFIA3 expression data in the Cancer Genome Atlas (TCGA) datasets and the clinical data of TNBC patients were downloaded. The relationship between PPFIA3 expression and clinicopathological factors as well as the prognosis were analyzed. By establishing co-expression networks, the gene co-expression datasets of PPFIA3 was analyzed by KEGG. Result The expression of PPFIA3 in malignant tumor tissues was higher than that in adjacent normal tissues (P〈0.01); and it was higher in patients with stage T3.4 than those with stage T1.2 disease and those without metastasis (P=0.025, 0.027). Survival analysis showed that patients with high PPFIA3 expression had shorter median survival time than those with low PPFIA3 expression (P=0.009). In KEGG signaling pathway enrichment analysis, it was proved that co-expressed genes of PPFIA3 participated in tumor necrosis factor (TNF), mitogen -activated protein kinases (MAPK), mechanistic target of rapamycin kinase (MTOR) and amino sugar and nucleotide sugar metabolism signaling pathway, etc. Conclusion PPFIA3 may be involved in the proliferation and metastasis of TNBC cells through signaling pathways such as TNF, MAPK, MTOR. High expression of PPFIA3 predicts poor prognosis for patients with TNBC. The expression of PPFIA3 might serve as an effective biomarker for predicting the metastasis and prognosis of TNBC patients.
