摘要
目的探讨促分裂原活化的蛋白激酶激酶(MEK)和胞外信号调节激酶(ERK)在肾癌骨转移患者中的表达及临床意义。方法回顾性分析80例原发性肾癌患者的临床资料,采用免疫组织化学法检测肾癌原发灶、癌旁组织及骨转移灶中MEK和ERK蛋白的表达水平,分析MEK和ERK蛋白表达水平在肾癌原发灶和骨转移灶中的相关性。结果有骨转移的肾癌原发灶和无骨转移的肾癌原发灶中MEK蛋白的阳性表达率分别为33.33%和23.53%,ERK蛋白的阳性表达率分别为41.67%和29.41%,均高于癌旁组织(P<0.05);有骨转移的肾癌原发灶和无骨转移的肾癌原发灶中MEK和ERK蛋白的阳性表达率比较,差异均无统计学意义(P>0.05);不同病理类型肾癌原发灶中MEK和ERK蛋白的阳性表达率比较,差异均无统计学意义(P>0.05);骨转移灶中MEK和ERK蛋白的阳性表达率分别为83.33%和91.67%,均高于有骨转移和无骨转移的肾癌原发灶(P<0.05);无骨转移的肾癌原发灶中MEK和ERK蛋白表达呈正相关(rs=0.707,P<0.05),有骨转移的肾癌原发灶中MEK和ERK蛋白表达也呈正相关(rs=0.897,P<0.05)。结论肾癌骨转移灶中MEK和ERK蛋白的阳性表达率均高于原发灶,MEK和ERK蛋白可能参与了肾癌骨转移过程,值得进一步研究。
Objective To investigate the expression and clinical significance of MEK and ERK protein in patients with bone metastasis of renal carcinoma.Method The clinical data of 80 patients with primary renal carcinoma were analyzed retrospectively, and the expression levels of MEK and ERK protein in renal carcinoma tissues, parcacancerous tissues and bone metastases tissues were detected by immunohistochemistry. The correlation between the expression levels of MEK and ERK protein in bone metastases tissues and renal carcinoma tissues was analyzed. Result The positive ex- pression rates of MEK protein in primary tumors with bone metastasis and without bone metastasis were 33.33% and 23.53%, the positive expression rates of ERK protein were 41.67% and 29.41% respectively, which were all significantly higher than those in paacancerous tissues (P〈0.05). There was no significant difference in the positive expression rate of MEK and ERK protein in the primary tumors with or without bone metastasis (P〉0.05). There were no significant difference in the positive expression rate of MEK and ERK protein between different pathological types of renal carcinoma (P〉 0.05). The positive expression rates of MEK and ERK protein in bone metastasis tissues were 83.33% and 91.67%, which were significantly higher than those in primary tumors with and without bone metastasis (P〈0.05). There was a positive correlation between the expression of MEK and ERK protein in the primary tumor of renal carcinoma without bone metastasis (rs=0.707, P〈0.05), and there was also a positive correlation between the expression of MEK and ERK protein in primary tumor of renal carcinoma with bone metastasis (rs=0.897, P〈0.05). Conclusion The positive expression rates of MEK and ERK protein in the bone metastases of renal carcinoma are both obviously higher than those of the primary tu- mors, which may be involved in the process of bone metastasis, and it is worth further study.
