摘要
以马铃薯基因数据库中基因ID为PGSC0003DMT400005378 (落酸受体PYR1基因)的核苷酸序列作为原始序列,用在线比对软件NCBI进行Blast比对其在拟南芥、番茄、烟草等物种中同源序列,以同源性为100%的核苷酸序列(GenBank accession No. XM_006361126.2)进行生物信息学分析,并作为模板序列设计引物,以马铃薯栽培品种‘陇薯3号’无菌试管薯为实验材料,用RT-PCR的方法获得StPYR1基因的cDNA序列,构建了其过表达载体pCPB-StPYR1,取得的结果可为研究StPYR1基因在马铃薯块茎休眠调控中的作用提供基础。
The nucleotide sequences of gene ID PGSC0003DMT400005378 (abscisic acid receptor PYR1 gene) from the potato genome databases was as original sequence, homologous sequences were compared among A rabidopsis thaliana, tomato, tobacco and other species using online software Blast of NCBI. Bioinformatics analysis was performed using 100% homologous sequences (GenBank accession No. XM 006361126.2), which was used to design primers as a template sequence. The sterile microtubers of potato cultivar ‘Longshu 3' was used as experimental materials. The eDNA sequence of StPYR 1 gene was obtained using RT-PCR method. The over-expression vector pCPB-StPYR1 was constructed. The result could provide basis for studying the role ofStPYR1 gene in regulating potato tuber dormancy.
