摘要
建立测定大鼠血浆中含葛根素量的方法,并应用于葛根素在大鼠体内的药代动力学研究.使用HPLC检测葛根素在大鼠血浆中的血药质量浓度,并应用Kinetica软件对葛根素的药代动力学参数进行非房室模型拟合.结果表明所建立的方法精密度、稳定性均良好,方法回收率和萃取回收率均符合药代动力学要求.葛根素在大鼠体内药代动力学参数为:C_(max)=(1 569. 67±216. 67) ng/mL,T_(max)=(0. 22±0. 075) h,AUC=(1 647. 87±108. 76)(h)*(ng/m L),t_(1/2)=(1. 61±0. 11) h.本实验明确了葛根素口服给药在大鼠体内的药代动力学情况,为药物的相互作用及临床合理用药提供参考数据.
To establish a method for determination of puerarin content in rat plasma, whichapply to study pharmacokinetics of puerarin in rat. Puerarin content in rat plasma was detec-ted by HPLC, and the phamlacokinetic parameters of puerarin was fitted with Kinetica. Theresults showed that the method was good in precision and stability, and the method recovecyand extraction recovery were in line with the phamlacokinetic requirements. The pharmacoki-netic parameters were Cmax = (1 569.67±216.67) ng/mL, Tmax= (0.22±0. 075) h, AUC= (1 647.87 ±108.76) (h) * (ng/mL), t1/2 = (1.61 ±0.11) h. In this paper, the phar-macokinetics of puerarin was given in rats, and reference data were provided for the drug -drug interaction and clinical rational drug.
