糖氧剥离对海马神经元Furin、BDNF表达水平的影响

摘要 目的探讨海马神经元在糖氧剥离时原蛋白转化酶Furin是否介导了脑源性神经营养因子(BDNF)表达水平的变化。方法通过OGD模拟缺血缺氧诱导海马神经元,采用免疫印迹观察OGD诱导后不同时间点BDNF、Furin动态表达水平变化。结果海马神经元经OGD诱导后再灌注12h内BDNF表达水平下调约30%(P<0.01);再灌注24hBDNF表达水平下调50%(P<0.01);再灌注48h其下降约70%,72h下调70%;与对照组比较有明显差异(P<0.001)。Furin表达水平呈进行性升高,即再灌注12h内Furin表达水平上调约1.3倍(P>0.05);再灌注24h时Furin表达水平上调2.1倍(P<0.001);再灌注48h其上调2.5倍,72h上调3.14倍;与对照组比较有明显差异(P<0.001)。结论经OGD诱导的海马神经元BDNF表达水平随再灌注时间呈进行性下降而Furin表达水平呈逐步上调。这一现象提示糖氧剥离时Furin未介导海马神经元内BDNF的细胞内分泌过程,在缺血缺氧时BDNF复杂的酶切过程有待进一步研究证实。 Objective To explore the effect of oxygen-glucose deprivation on Furin and BDNF expression levels of hippocampus neuron.Methods In vitro primary culture was conducted on the neurons in hippocampus area of newborn SD rats within 24 hours.The cultured neuron was induced by oxygen—glucose deprivation(OGD).Protein was extracted in 0 h,12 h,24 h,48 hand 72 hafter OGD,and Western Blotting was used to test mBDNF and Furin expression levels at different time points.Results Compared with the control group,mBDNF gradually decreased,30%in 12 h,50%in 24 h,70% in 48 hand 70% in 72 h;while furin gradually increased,in 1.3 fold in 12 h,2.1 fold in 24 h,2.5 fold in 48 hand 3.14 fold in 72 hafter OGD.Conclusion With the injury of hippocampal neuron,OGD could result in the change of the mBDNF and Furin expression levels and Furin didn't mediate the change of the BDNF expression level.The mechanism of the downregulation of BDNF would be explored further.
